Inhibition of RhoA/MRTF-A signaling alleviates nucleus pulposus fibrosis induced by mechanical stress overload

Mengxiong Song et al.

Summary: The study found that collagen I, MMP-12, and CTGF were significantly upregulated in IDD patients, while aggrecan and collagen II were downregulated. Overloaded strain increased the nuclear translocation of MRTF-A and changed the morphology of NPCs. Additional experiments showed upregulation of RhoA, ROCK2, MRTF-A, SRF, MMP-12, and CTGF, and downregulation of aggrecan and collagen II under overload strain. The RhoA/MRTF-A pathway inhibitor CCG-1423 reversed strain-induced fibrosis.

A High Body Mass Index and the Vacuum Phenomenon Upregulate Pain-Related Molecules in Human Degenerated Intervertebral Discs

Masayuki Miyagi et al.

Summary: This study investigates the contribution of mechanical stress and axial loading to the development of discogenic low back pain. By analyzing clinical findings, radiographical findings, and pain-related molecules expression in human degenerated intervertebral discs, the study finds that axial loading and mechanical stress upregulate CGRP and mPGES-1, potentially leading to chronic discogenic low back pain.

Painful intervertebral disc degeneration and inflammation: from laboratory evidence to clinical interventions

Feng-Juan Lyu et al.

Summary: Low back pain is a common musculoskeletal disorder with inflammation playing a crucial role in disc degeneration and pain. Various animal models and signaling pathways related to painful disc degeneration have been discussed, along with potential drug candidates to suppress discogenic LBP by targeting inflammation.
