Proteomic Assessment of Extracellular Vesicles from Canine Tissue Explants as a Pipeline to Identify Molecular Targets in Osteosarcoma: PSMD14/Rpn11 as a Proof of Principle

Anita K. Luu et al.

Summary: Osteosarcoma (OS) is a highly malignant bone tumor with limited improvements in treatment. Understanding the molecules contributing to OS biology can lead to the discovery of novel therapies. This study demonstrates the potential of extracellular vesicles (EVs) in revealing protein signatures related to OS and highlights a specific protein, PSMD14/Rpn11, as a potential prognostic marker and therapeutic target for OS.

Recent and Ongoing Research into Metastatic Osteosarcoma Treatments

Michael A. Harris et al.

Summary: The survival rate for metastatic osteosarcoma has remained stagnant for decades despite the introduction of chemotherapy as a treatment option. Efforts to identify potential therapies through animal studies and clinical trials have had limited success in improving survival outcomes for these patients.

CD44 Contributes to the Regulation of MDR1 Protein and Doxorubicin Chemoresistance in Osteosarcoma

Monserrat Gerardo-Ramirez et al.

Summary: Osteosarcoma is the most common type of pediatric bone tumor. Research shows that CD44 plays an important role in drug resistance of osteosarcoma cells, and abnormal expression of CD44 leads to molecular alterations.

Analyzing BMP2, FGFR, and TGF Beta Expressions in High-Grade Osteosarcoma Untreated and Treated Autografts Using Proteomic Analysis

Rashmi Madda et al.

Summary: Biological reconstruction techniques have greatly improved the treatment of high-grade osteosarcoma patients by using liquid nitrogen and irradiation processes to remove tumors and reconstruct autografts. This proteomic study focuses on the expressions of growth factor family proteins in untreated and treated autografts, which play a crucial role in bone union, remodeling, and regeneration. The study found that these protein expressions were significantly higher in treated autografts, suggesting that the increased growth factor protein expression may contribute to bone regeneration healing and reduce the recurrence and spread of high-grade sarcoma.

Challenges of Systemic Therapy Investigations for Bone Sarcomas

Kenji Nakano

Summary: Bone sarcoma, a rare component of malignant solid tumors, presents various histological types and genomic mutations. Targeted drug investigations for bone sarcomas have not progressed like those for other solid tumors, with challenges in patient recruitment, especially among pediatric and young adult patients.

15d-PGJ2 Promotes ROS-Dependent Activation of MAPK-Induced Early Apoptosis in Osteosarcoma Cell In Vitro and in an Ex Ovo CAM Assay

Mateja Mikulcic et al.

Summary: The study revealed that 15-Deoxy-Delta(12,14)-prostaglandin J(2) (15d-PGJ(2)) exhibits significant anti-tumor effects on osteosarcoma cells by promoting apoptosis, increasing intracellular ROS levels, and activating the MAPK pathway, offering a promising natural compound to interfere with osteosarcoma tumor growth.

Jir-You Wang et al.

Summary: Osteosarcoma is a highly malignant musculoskeletal tumor commonly seen in various age groups, with a survival rate of 70-80%. ER alpha plays a significant role in osteosarcoma pathogenesis and chemosensitivity, with potential as a target for current treatments and a prognostic factor for chemotherapy response.

Extracellular Vesicles in Osteosarcoma: Antagonists or Therapeutic Agents?

Viviana De Martino et al.

Summary: Osteosarcoma is a skeletal tumor that predominantly affects children and adolescents, often metastasizing to the lungs. Current treatment approaches involve surgery and chemotherapy, but drug resistance and side effects necessitate the search for new therapeutic strategies. Extracellular vesicles have sparked interest as potential diagnostic biomarkers and drug delivery vehicles for osteosarcoma, offering a promising avenue for treatment advancement.
