Function of ceramide transfer protein for biogenesis and sphingolipid composition of extracellular vesicles

Summary: The ceramide transport protein CERT plays a crucial role in regulating the composition and biogenesis of extracellular vesicles (EVs). It mediates a non-vesicular transport of ceramide between the endoplasmic reticulum and multivesicular endosome, and forms a complex with ESCRT-I protein. Inhibition of ceramide biosynthesis reduces the formation of CERT-Tsg101 complex. Overexpression of CERT increases EV secretion, while its inhibition reduces EV formation and the concentration of ceramides and sphingomyelins in EVs.

HRS phosphorylation drives immunosuppressive exosome secretion and restricts CD8+ T-cell infiltration into tumors

Summary: Inadequate infiltration of CD8(+) T cells into tumors is associated with poor response to anti-PD-1 therapy. This study reveals that phosphorylation of HRS restricts tumor infiltration of CD8(+) T cells by promoting the loading of immunosuppressive exosomes containing PD-L1. Inhibition of HRS phosphorylation enhances the efficacy of anti-PD-1 treatment in mouse melanoma models. These findings suggest that blocking HRS phosphorylation could be a potential strategy to improve the effectiveness of cancer immunotherapy.
