Pulsed Electric Field Ablation of Esophageal Malignancies and Mitigating Damage to Smooth Muscle: An In Vitro Study

Action spectra and mechanisms of (in) efficiency of bipolar electric pulses at electroporation

Summary: This study aimed to achieve the maximum cancellation of electroporation by quantifying the impact of pulse shape, duration, number, and repetition rate on a broad range of electric field strengths. The addition of an opposite polarity phase reduced cell membrane permeabilization, with the strongest cancellation achieved by adding a 50% second phase to 600-ns pulses delivered at a high repetition rate. Strobe photography revealed that the temporal summation of polarization enhanced electroporation for unipolar pulses, while no summation was observed for bipolar pulses.

Summary: Electroporation, particularly with nanosecond pulses, is an efficient technique for creating nanometer-size membrane lesions without toxins. Membrane restoration involves both fast, calcium-independent resealing and slow, calcium-dependent processes. Additional calcium can advance resealing and reduce dye uptake, while blocking calcium does not prevent full membrane recovery.

Additionally, restricting the frequency of bipolar nsPEF bursts to the refractory period after nerve excitation can minimize the number of neuromuscular reactions.
