Antiviral Responses of Tissue-resident CD49a1 Lung Natural Killer Cells Are Dysregulated in Chronic Obstructive Pulmonary Disease

Grace E. Cooper et al.

Summary: This study investigates the phenotype and antiviral responses of tissue-resident natural killer (trNK) cells in murine chronic obstructive pulmonary disease (COPD) and human COPD lung tissue. The results show that CD49a(+) NK cells in the lung have a distinct phenotype compared to circulating NK cells and their proportions increase with worsening disease. The study suggests that trNK cell function is altered in cigarette smoke-induced disease and may contribute to excessive inflammation during viral exacerbations of COPD.

Targeting intercellular adhesion molecule-1 (ICAM-1) to reduce rhinovirus-induced acute exacerbations in chronic respiratory diseases

Shakti D. Shukla et al.

Summary: Asthma and COPD are major causes of global mortality and morbidity, with HRV being implicated in exacerbations of both diseases. ICAM-1 plays a crucial role in facilitating HRV adhesion and transmission to the respiratory system, with smoking leading to up-regulation of ICAM-1 and increased susceptibility to viral infections in individuals with chronic lung diseases. Therapeutic strategies focusing on blocking ICAM-1 are being explored to prevent viral adhesion and exacerbations of COPD.

Gut bacteria alleviate smoking-related NASH by degrading gut nicotine

Bo Chen et al.

Summary: Tobacco smoking is positively correlated with non-alcoholic fatty liver disease (NAFLD), but the underlying mechanism is unclear. This study found that nicotine accumulates in the intestine during tobacco smoking and activates intestinal AMPK alpha. The gut bacterium Bacteroides xylanisolvens is identified as an effective nicotine degrader. These findings highlight the role of intestinal nicotine accumulation in NAFLD progression and the presence of an endogenous bacterium in the human intestine that can metabolize nicotine.

Summary: The prevalence of chronic immune and metabolic disorders is increasing rapidly, with significant associations found between alterations in the microbiome, diet, and mucous barriers in the onset and progression of these diseases. The interplay between diet, microbiome, and mucous barrier has gained attention in understanding major immune/metabolic disorders.

A monoclonal antibody to Siglec-8 suppresses non-allergic airway inflammation and inhibits IgE-independent mast cell activation

Julia Schanin et al.

Summary: Studies have shown that therapeutic treatment with anti-Siglec-8 monoclonal antibody can inhibit mast cell activation, reduce recruitment of immune cells and airway inflammation, and alleviate lung fibrosis in non-allergic disease models. Additionally, anti-Siglec-8 treatment can also suppress neutrophil influx and cytokine production by inhibiting mast cell activation.
