CBD activation of TRPV1 induces oxidative signaling and subsequent ER stress in breast cancer cell lines

Amy de la Harpe et al.

Summary: ER stress is an imbalance between protein-folding load and ER capacity, which can be re-established through UPR. Studies suggest that CBD may induce ER stress via Ca2+ influx through the TRPV1 receptor, increasing ROS levels and disrupting protein folding.

Regulation of calcium homeostasis and flux between the endoplasmic reticulum and the cytosol

Lea Daverkausen-Fischer et al.

Summary: Maintaining proper Ca2+ concentration in ER is crucial for maintaining its oxidizing environment and luminal ATP levels. Ca2+ influx into ER is enabled by a reductive shift while Ca2+ leakage occurs through the Sec61 translocon. Dysregulated Ca2+ metabolism can induce the unfolded protein response in cells.

Summary: This study demonstrates the neuroprotective effects of cocoa extract and its purified fractions in a cellular model of Parkinson's disease. The cocoa extract targets the PERK sensor, showing significant antioxidant and antiapoptotic activities. Additionally, cocoa inhibits the accumulation of alpha-synuclein in the cells.

Calcium channel TRPV6 promotes breast cancer metastasis by NFATC2IP

Xiang Xu et al.

Summary: The upregulation of calcium channel TRPV6 is associated with poor prognosis in breast cancer by promoting invasion and metastasis, making it a potential target for therapy. TRPV6 expression is increased in metastatic breast cancers, accelerating primary breast cancer cell migration. Mechanistically, TRPV6 activates NFATC2 through NFATC2IP phosphorylation, leading to increased breast cancer metastasis via upregulation of ADAMTS6 expression.

The molecular mechanism and functional diversity of UPR signaling sensor IRE1

Samirul Bashir et al.

Summary: The endoplasmic reticulum is responsible for protein folding and maturation, but can be affected by various stress conditions triggering the Unfolded Protein Response. This response involves the activation of sensors like IRE1 which signal to restore cellular homeostasis, and IRE1 can switch its substrate specificity to cotransnationally transported mRNAs in a process known as Regulated IRE1 Dependent Decay (RIDD).

The aftermath of the interplay between the endoplasmic reticulum stress response and redox signaling

Kashi Raj Bhattarai et al.

Summary: The endoplasmic reticulum (ER) plays crucial roles in protein synthesis and folding in eukaryotic cells. The unfolded protein response (UPR) is triggered by disturbances in ER function, aiming to restore cellular homeostasis but possibly leading to apoptotic signaling under prolonged stress. The complex interactions between ER stress, redox imbalance, and oxidative stress can have profound negative impacts on cellular physiology and targeting these stress mechanisms may be a promising approach for disease treatment.

PERK Pathway and Neurodegenerative Disease: To Inhibit or to Activate?

Talya Shacham et al.

Summary: With the extension of life span, there is an increasing burden of late-onset neurodegenerative diseases, for which effective treatments are lacking. Strategies to inhibit cellular toxicity and restore homeostasis are drawing increasing attention as gene therapy approaches face difficulties in tackling these diseases. Both inhibition and activation of the PERK pathway have shown benefits in disease models, but the complexity of the pathway and its effects on cell survival or death have led to conflicting results.

Crosstalk Between ER Stress, Autophagy and Inflammation

Sandhya Chipurupalli et al.

Summary: The endoplasmic reticulum (ER) is crucial for protein synthesis, folding, sensing cellular stress and maintaining cellular homeostasis. The unfolded protein response (UPR) expands protein folding capacity, reduces protein synthesis, enhances ER-associated protein degradation (ERAD), and amplifies cytokine-mediated inflammatory responses. UPR also activates autophagy, which limits inflammation and provides cyto-protection by attenuating ER-stress.

Structural and Functional Significance of the Endoplasmic Reticulum Unfolded Protein Response Transducers and Chaperones at the Mitochondria-ER Contacts: A Cancer Perspective

Giuseppina Amodio et al.

Summary: The ER has emerged as a key coordinator of cellular homeostasis due to its connection to almost all intracellular organelles. Intense crosstalk between the ER and mitochondria at MERCs regulates fundamental cellular processes like cell fate decision, metabolism, and proteostasis, impacting tumorigenesis and therapeutic response of cancer cells. Recent studies show that UPR machinery components at MERCs have multifaceted roles related to controlling calcium fluxes to mitochondria, influencing cell survival and death.
