Antifibrotic effect of brown algae-derived fucoidans on osteoarthritic fibroblast-like synoviocytes

Summary: Synovial fibrosis is a pathological process that causes joint pain and stiffness. Fucoidans, a type of sulfated polysaccharides found in brown algae, have shown promise as therapeutic agents. In vitro studies have found that fucoidans inhibit the differentiation of fibroblast-like synoviocytes into myofibroblasts and restore apoptosis. The protective effects of these sulfated polysaccharides are mediated by upregulation of nitric oxide production and modulation of the TGF-beta/smad pathway. These findings suggest that fucoidans could be used as therapeutic compounds for fibrotic processes in rheumatic pathologies.

Reduced Levels of H2S in Diabetes-Associated Osteoarthritis Are Linked to Hyperglycaemia, Nrf-2/HO-1 Signalling Downregulation and Chondrocyte Dysfunction

Summary: Different findings suggest that type 2 diabetes is a risk factor for osteoarthritis, but the underlying mechanisms are unclear. This study examined the role of hydrogen sulphide (H2S) in the development of diabetes-related osteoarthritis. The results showed a reduction in H2S synthesis in cartilage from diabetic patients and cells under glucose stress. H2S was found to be involved in the Nrf-2/HO-1 signaling pathway and its impairment led to chondrocyte catabolic responses. Therefore, using exogenous H2S may be beneficial for the treatment of diabetes-related osteoarthritis.

Fucoidan, a class of sulfated polysaccharides, has various pharmacological properties and can protect against oxidative stress. This study isolated fucoidan from four brown seaweeds and investigated its protective effect against high glucose-induced oxidative stress in heart cells. The results showed that one of the isolated fucoidan had higher antioxidant activity and could protect against oxidative stress and preserve cellular function in the heart cells.
