ANO1: More Than Just Calcium-Activated Chloride Channel in Cancer

Saisai Guo et al.

Summary: This article reviews the role of ANO1 in various malignant tumors, including replication, proliferation, invasion, and apoptosis. The expression of ANO1 is controlled by various molecules and stimuli, with different regulatory mechanisms in different tumor cells. Studies have shown that ANO1 exerts its effects through multiple pathways and signaling pathways, and has functions in resisting apoptosis and promoting immune escape. Further research on the channels and protein activity mechanism of ANO1, as well as the development of ANO1 inhibitors, is of great significance for revealing its tumor-promoting mechanism.

Cinobufagin Exerts Anticancer Activity in Oral Squamous Cell Carcinoma Cells through Downregulation of ANO1

Sungwoo Jo et al.

Summary: The study identified cinobufagin as a novel regulator that can downregulate the expression of ANO1 in oral squamous cell carcinoma cells CAL-27, leading to inhibition of cell proliferation, migration, and induction of apoptosis. These findings suggest that downregulation of ANO1 by cinobufagin may be a potential mechanism for its anticancer effects in OSCC.

The Ca2+-activated chloride channel ANO1/TMEM16A: An emerging therapeutic target for epithelium-originated diseases?

Yani Liu et al.

Summary: ANO1 gene encodes a Ca2+ activated Cl- channel critical for physiological functions, with abnormal expression or dysfunction linked to various diseases. The lack of specific modulators hinders the validation of ANO1 as a therapeutic target.

Towards resolving the enigma of the dichotomy of resveratrol: cis- and trans-resveratrol have opposite effects on TyrRS-regulated PARP1 activation

Megha Jhanji et al.

Summary: Resveratrol can extend replicative lifespan and have health benefits, but may also pose risks such as nerve degeneration. Its structural similarity to tyrosine allows it to act as an antagonist to tyrosyl-tRNA synthetase, inducing a protective stress response.

Diethylstilbestrol, a Novel ANO1 Inhibitor, Exerts an Anticancer Effect on Non-Small Cell Lung Cancer via Inhibition of ANO1

Yohan Seo et al.

Summary: Non-small cell lung cancer (NSCLC) is a leading cause of cancer-related mortality, and ANO1 plays a crucial role in its treatment. This study identified DES as a selective ANO1 inhibitor, which effectively reduces ANO1 channel activity and protein levels, inhibiting the survival and migration of NSCLC cells.
