The pleiotropic peroxisome proliferator activated receptors: Regulation and therapeutics

Gargi Dixit et al.

Summary: PPARs play a crucial role in regulating metabolic events in our body, with their agonists and antagonists holding therapeutic significance. Understanding the molecular mechanisms of different PPAR isotypes in health and disease is essential for maximizing their therapeutic potential. Insights into the multifaceted therapeutic effects and underlying mechanisms of PPAR agonists and antagonists will guide future efforts in identifying novel molecules for metabolic, inflammatory, and immunotherapeutic diseases.

Summary: The study evaluated the effects of bimatoprost on orbital fat in rats, demonstrating a significant decrease in adipocytes cell count in the treated orbits and a complete absence of fat cells in some of the treated orbits. Bimatoprost may be an effective treatment for inactive thyroid eye disease.

Mitophagy: Molecular Mechanisms, New Concepts on Parkin Activation and the Emerging Role of AMPK/ULK1 Axis

Roberto Iorio et al.

Summary: Mitochondria are critical organelles for cellular energy balance and cell death. Mitophagy, the selective removal of dysfunctional mitochondria by autophagy, plays important roles in maintaining mitochondrial health. Both impaired and excessive mitophagy are involved in various ageing-associated diseases. Recent advances have explored the molecular mechanisms and signaling pathways that regulate mitophagy, including the early role and spatial specificity of the AMPK/ULK1 axis.

Ocular benzalkonium chloride exposure: problems and solutions

Michael H. Goldstein et al.

Summary: Benzalkonium chloride (BAK) is a commonly used preservative in eye medications, but long-term exposure can cause cytotoxic damage to the ocular surface, resulting in symptoms of ocular surface disease. There are multiple strategies available to minimize or eliminate BAK exposure, including alternative preservatives and preservative-free formulations.

Synergistic Activity of Ketoconazole and Miconazole with Prochloraz in Inducing Oxidative Stress, GSH Depletion, Mitochondrial Dysfunction, and Apoptosis in Mouse Sertoli TM4 Cells

Sabrina Petricca et al.

Summary: Triazole and imidazole fungicides, as emerging pollutants with endocrine-disrupting properties, have been found to have adverse effects on male reproductive cells. This study specifically demonstrates that ketoconazole and miconazole, alone or in combination with prochloraz, significantly impair the functions of Sertoli cells and cause oxidative stress. However, fluconazole shows no cytotoxic effects.

Molecular regulation of neuroinflammation in glaucoma: Current knowledge and the ongoing search for new treatment targets

Gulgun Tezel

Summary: Neuroinflammation plays a significant role in the neurodegenerative processes of glaucoma, with glial cells and peripheral immune cells causing toxicity to retinal ganglion cells. The interaction between glial cells and immune cells is crucial in amplifying neuroinflammation and determining the neurodegenerative outcome. Inhibition of neurodegenerative inflammation may provide a broad treatment strategy for rescuing retinal ganglion cell integrity and function, while targeting both inflammatory toxicity and mitochondrial dysfunction.

Ocular Tolerability of Bimatoprost 0.1 mg/mL Preservative-Free versus Bimatoprost 0.1 mg/mL with Benzalkonium Chloride or Bimatoprost 0.3 mg/mL Preservative-Free in Patients with Primary Open-Angle Glaucoma

Mariaelena Filippelli et al.

Summary: This study evaluated the therapeutic switch from Bimatoprost formulations with or without benzalkonium chloride (BAK) to a preservative-free formulation and its impact on eye surface conditions and intraocular pressure (IOP) in glaucoma patients. The results showed that Bimatoprost 0.1 mg/mL preservative-free had better tolerability and improved eye surface conditions compared to other formulations, but had slightly inferior IOP control.

Multi-Target Effects of ss-Caryophyllene and Carnosic Acid at the Crossroads of Mitochondrial Dysfunction and Neurodegeneration: From Oxidative Stress to Microglia-Mediated Neuroinflammation

Roberto Iorio et al.

Summary: Inflammation and oxidative stress are closely related in chronic diseases, and targeting inflammatory pathways may be a potential therapeutic strategy. Phytochemicals extracted from edible plants, such as ss-caryophyllene and carnosic acid, have demonstrated anti-inflammatory, antioxidant, and neuroprotective effects in various experimental models. These compounds have the potential to maintain mitochondrial homeostasis and ameliorate glia-mediated neuroinflammation in the treatment of neurodegenerative diseases.

Lactobacillus sakei Pro-Bio65 Reduces TNF-α Expression and Upregulates GSH Content and Antioxidant Enzymatic Activities in Human Conjunctival Cells

Roberto Iorio et al.

Summary: The study investigates the regulatory effects of Lactobacillus sakei pro-Bio65 lysate on human conjunctival epithelial cells, showing that it does not affect cell proliferation but induces low levels of intracellular ROS and changes in mitochondrial membrane potential. Additionally, it upregulates antioxidant defense efficiency and reduces TNF-alpha expression, suggesting the potential application of L.SK paraprobiotic in promoting eye health and preventing oxidative stress-related conditions.
