Evaluation of Approach to a Conspecific and Blood Biochemical Parameters in TAAR1 Knockout Mice

Ilya S. Zhukov et al.

Summary: This study evaluated the sexual behavior of TAAR1 knockout and wild-type mice using a sexual incentive motivation test, and measured testosterone and other biochemical parameters in their blood. The results showed that the lack of TAAR1 does not significantly affect sexual motivation and routine lipid and metabolic blood biochemical parameters.

Trace Amine Associate Receptor 1 (TAAR1) as a New Target for the Treatment of Cognitive Dysfunction in Alzheimer's Disease

Damiana Leo et al.

Summary: Approximately 27 million people worldwide are affected by Alzheimer's disease (AD), mainly caused by the deposition of Aβ. The TAAR1 agonist RO5256390 may have potential in improving cognitive impairments induced by Aβ.

Trace Amine-Associated Receptor 1 as a Target for the Development of New Antipsychotics: Current Status of Research and Future Directions

Joshua T. Kantrowitz

Summary: This review assesses the role of trace amine-associated receptor 1 (TAAR-1) in schizophrenia and the potential of TAAR-1 modulators as novel-mechanism antipsychotics. It includes an overview of TAAR-1 physiology and pathophysiology in schizophrenia, interaction with other neurotransmitter systems, and a review of investigational TAAR-1 compounds. Preliminary results for ulotaront show positive effects on overall schizophrenia symptoms without significant tolerability concerns, suggesting the potential for expanding the definition of antipsychotics beyond the D2R target.

Therapeutic Potential of TAAR1 Agonists in Schizophrenia: Evidence from Preclinical Models and Clinical Studies

Nina Dedic et al.

Summary: TAAR1 has emerged as a promising therapeutic target for neuropsychiatric disorders due to its ability to modulate neurotransmission, particularly in regulating dopamine. Agonist compounds targeting TAAR1 have shown potential as treatments for schizophrenia and other disorders. Clinical trial data, along with preclinical findings, provide a rationale for further exploration and development of this new pharmacological class for treating related disorders.

Genetically identified amygdala-striatal circuits for valence-specific behaviors

Xian Zhang et al.

Summary: Researchers have discovered two classes of 'hardwired' neurons in the BLA, positive-valence and negative-valence neurons, which express Fezf2 and have distinct connectivity. These neurons drive punishment avoidance and reward seeking behaviors by receiving inputs from separate sensory and limbic areas, and conveying information to the nucleus accumbens and olfactory tubercle.

Dopamine promotes aggression in mice via ventral tegmental area to lateral septum projections

Darshini Mahadevia et al.

Summary: The authors demonstrate that terminal activity of dopaminergic neurons projecting from the ventral tegmental area to the lateral septum plays a key role in promoting aggression in mice. Dopamine acts on D2 receptors to inhibit GABAergic neurons within the lateral septum, and septal D2 signaling is essential for ventral tegmental area dopamine activity to promote aggression.
