Timo D. Mueller et al.

Summary: Enormous progress has been made in managing diseases associated with excess body weight, but treatment of obesity itself remains challenging. By summarizing the history and advancements in anti-obesity medications, researchers are now pursuing next-generation AOMs that could safely and effectively achieve weight loss.

Contribution of markers of adiposopathy and adipose cell size in predicting insulin resistance in women of varying age and adiposity

Eve-Julie Tremblay et al.

Summary: The dysfunctions of adipose tissue are associated with cardiometabolic risk in women, and adipocyte cell size and the adiponectin/leptin ratio play important roles in lipid-lipoprotein levels and insulinemia. The adiponectin/leptin ratio is a significant predictor of fasting insulin levels and HOMA-IR. Leptin levels are a better predictor of insulin resistance than adipocyte cell size and the adiponectin/leptin ratio.

Sex and Species Differences in the Development of Diet-Induced Obesity and Metabolic Disturbances in Rodents

Ivana Maric et al.

Summary: The prevalence and health consequences of obesity differ between men and women. Research shows that male and female rodents respond differently to high-fat diet (HFD) challenge, with female rodents displaying a higher preference for HFD but experiencing a delay in weight gain and fewer metabolic complications compared to males. These sex-based differences are divergent in rats and mice, with male SD rats being a better model for the lower prevalence of overweight in women, while C57BL/6 mice may better represent the increased prevalence of morbid obesity in women.

Impaired Suppression of Glucagon in Obese Subjects Parallels Decline in Insulin Sensitivity and Beta-Cell Function

Xi Chen et al.

Summary: The study revealed that there is a relationship between plasma glucagon levels and insulin sensitivity and insulin secretion in obese individuals. As glucose intolerance worsens, the insulin-mediated suppression of glucagon secretion is impaired, which corresponds to the severity of insulin resistance and beta-cell dysfunction.

Summary: Studies on rats fed high-fat/cafeteria diets show an increase in brown adipose tissue thermogenesis, though some studies lack details on diet composition and housing temperature. Most studies superficially investigate thermogenesis, focusing mainly on UCP1 expression.

An Explanation for the Adiponectin Paradox

Hans O. Kalkman

Summary: Adiponectin improves insulin sensitivity, but overnutrition can lead to decreased synthesis of adiponectin and reduced signal transduction, causing impairment in insulin sensitivity. Different treatments may be needed for hypoadiponectinemia and hyperadiponectinemia, despite both reflecting insulin resistance.

Diet-induced obesity in animal models: points to consider and influence on metabolic markers

Mariana de Moura e Dias et al.

Summary: Overweight and obesity are global public health problems that have increased considerably in prevalence. Diet-induced obesity animal models can replicate human overweight and obesity, with various protocols to lead to excess fat deposition. Key points for inducing obesity through diet include evaluating body weight, fat gain, inflammation, hormonal concentration, blood glycemia, lipid profile, liver health, and considering diet composition and calorie overconsumption for obesity development.

Metabolic Disturbances in Rat Sublines with Constitutionally Altered Serotonin Homeostasis

Maja Kesic et al.

Summary: This study found that rats with high central and peripheral serotonin levels developed obesity in the abdominal area, along with glucose and insulin intolerance. Despite elevated glucose and lipid levels in their blood, these rats did not show significant changes in leptin, resistin, and adipsin. Surprisingly, their adiponectin levels were increased in plasma but reduced in white adipose tissue. Additionally, a few metabolic genes in the high-serotonin rats showed differential expression in WAT compared to low-serotonin rats.

Single or combined ablation of peripheral serotonin and p21 limit adipose tissue expansion and metabolic alterations in early adulthood in mice fed a normocaloric diet

Enrica Saponara et al.

Summary: Identifying molecular factors such as serotonin and p21 that promote physiological weight gain and metabolic alterations provides potential targets for preventing obesity. Research in mice on a normal calorie diet revealed the importance of these molecules in adipose tissue expansion and systemic perturbations in glucose metabolism. Targeting serotonin and p21 could offer a pharmacologic approach to combat the global pandemic of obesity.
