Overexpression of NMNAT3 improves mitochondrial function and enhances antioxidative stress capacity of bone marrow mesenchymal stem cells via the NAD+-Sirt3 pathway

Tao Wang et al.

Summary: This study confirms that overexpressing NMNAT3 can improve mitochondrial function and resistance to stress-induced apoptosis in BMSCs. By increasing NAD+ levels, NMNAT3 can increase the activity of Sirt3 and decrease the acetylation levels of related proteins. This provides a new mechanism and approach for improving the survival rate and curative effect of BMSCs.

The Role of SIRT3 in Exercise and Aging

Lei Zhou et al.

Summary: The health benefits of regular exercise are well-established, but the molecular mechanisms behind these benefits are still debated. SIRT3, a key cell-signaling candidate, has been proposed to provide exercise-induced benefits. However, studies on the impact of exercise on SIRT3 abundance in cells have conflicting results. This narrative review analyzes the role of exercise-induced changes in SIRT3 in providing the health and aging benefits associated with regular physical activity, and raises important unanswered questions for future research.

SIRT3 is required for liver regeneration but not for the beneficial effect of nicotinamide riboside

Sarmistha Mukherjee et al.

Summary: This study reveals that the loss of mitochondrial deacetylase SIRT3 in hepatocytes impairs liver regeneration and worsens mitochondrial function after partial hepatectomy. While NAD precursor NR promotes liver regeneration, it is shown that neither SIRT3 nor SIRT1 is necessary for NR's enhancing effect. NR improves mitochondrial respiration in regenerating livers and increases oxygen consumption and glucose output in cultured hepatocytes, indicating a direct enhancement of mitochondrial metabolism as the mechanism for improved liver regeneration after NAD supplementation.

Honokiol attenuates lipotoxicity in hepatocytes via activating SIRT3-AMPK mediated lipophagy

Jingxin Liu et al.

Summary: The study found that honokiol can alleviate lipid accumulation by promoting SIRT3-AMPK-mediated autophagy, mainly targeting lipid droplets. Honokiol interacts hydrophobically with amino acid residues (PHE294, GLU323, and VAL324) and NAD(+). Additionally, honokiol improves mitochondrial function for enhanced lipolysis by reducing acetylated long-chain acyl-CoA dehydrogenase levels. In CDHFD-fed mice, honokiol treatment significantly prevented liver lipid accumulation, and co-treatment with the AMPK inhibitor Compound C almost abolished these effects.
