Inference of CRISPR Edits from Sanger Trace Data

David Conant et al.

Summary: ICE is a tool for analyzing CRISPR edits using Sanger data, providing a fast, accurate, and affordable way to assess the genotype after editing. It proposes potential editing outcomes and determines the data-supported results through regression analysis. ICE can analyze CRISPR experiments within days after transfection and produces accurate estimates of editing outcomes in various benchmarks and existing tools.

Temperature dependent in vitro binding and release of target DNA by Cas9 enzyme

Serene Rose David et al.

Summary: This study investigates the binding properties of Cas9 enzyme to specific target DNA at different temperatures and finds that Cas9 enzyme can efficiently bind and cleave DNA at temperatures as low as 4 degrees C. The study also shows that the cleaved DNA products remain bound to the Cas9 enzyme strongly and are released only at higher temperatures. These findings provide valuable insights for developing temperature-dependent applications of the CRISPR-Cas9 technology.
