Comparison of Whole Blood Cryopreservation Methods for Extensive Flow Cytometry Immunophenotyping

Summary: Fresh blood immunophenotyping by flow cytometry is the best method to study the human immune system, but sample quality is difficult to maintain in large studies. Our study compared the quality of immunophenotypic data obtained from fresh blood with those obtained after cryopreservation. The method using dimethyl sulfoxide solution produced results most similar to those with fresh samples.

Comprehensive evaluation of the effects of long-term cryopreservation on peripheral blood mononuclear cells using flow cytometry

Summary: This study used flow cytometry to investigate the effect of long-term cryopreservation on the quality of peripheral blood mononuclear cells (PBMCs), and found that long-term cryopreservation did not affect the recovery and viability of PBMCs, but led to a decrease in the number of innate immune cells and dynamic changes in the proportions of T cell subtypes. This study provides important evidence for the effective utilization of biobank resources.

An easy and reliable whole blood freezing method for flow cytometry immuno-phenotyping and functional analyses

Summary: This study presented an optimized protocol for rapid whole blood freezing with a specific solution, which allows effective preservation of various types of immune cells. The results showed a viability rate of over 75% for overall cells, with frequencies of major immune cell types comparable to fresh samples, albeit with a slight decrease in B cell frequencies.

Effects of long-term cryopreservation of PBMC on recovery of B cell subpopulations

Summary: After 12 months of cryopreservation of PBMC, there was a gradual decline in B cell viability and recovery, while no significant changes were observed in the proportional representation of B cell subpopulations. The functional response elicited remained comparable to that observed after short-term storage for one month.
