Could low α-N-acetylgalactosaminidase plasma concentration cause schizophrenia?

Summary: Using a neurodevelopmental approach, this study investigated the etiology of schizophrenia and found an enzyme deficiency at the cellular level. The results showed that schizophrenic patients had lower levels of α-N-acetylgalactosaminidase (α-NAGAL) compared to the control group, and there was a correlation between α-NAGAL levels and disease severity and age of onset. These findings support the neurodevelopmental hypothesis and suggest that α-NAGAL may play an important role in the etiology of schizophrenia.

Neuronal-Derived EV Biomarkers Track Cognitive Decline in Alzheimer's Disease

Summary: The hallmarks of Alzheimer's disease (AD) pathology are senile plaques containing amyloid-beta (A beta) and neurofibrillary tangles containing hyperphosphorylated tau. Additional pathologies often co-exist, whereas multiple pathogenic mechanisms are involved in AD, especially synaptic degeneration, which necessitate the need for synaptic integrity-related biomarkers alongside A beta- and tau-related biomarkers. Plasma neuron-derived Extracellular Vesicles (NDEVs) provide biomarkers related to A beta and tau and synaptic degeneration. Higher levels of NDEV A beta(42) were consistently associated with better cognitive status, memory, fluency, working memory, and executive function. Higher levels of NDEV synaptic integrity-related biomarkers were associated with better performance on executive function tasks. Our findings support the hypothesis that releasing A beta(42)-laden NDEVs may be an adaptive mechanism in AD.

Meprin β knockout reduces brain Aβ levels and rescues learning and memory impairments in the APP/lon mouse model for Alzheimer's disease

Summary: This study reports the generation of a transgenic mouse model of AD lacking the functional Mep1b gene (APP/lon x Mep1b(-/-)). The results showed that when meprin beta is absent, the levels of A beta 1-40 and 1-42 are reduced in APP/lon mice, and the deposition of N-terminally truncated A beta 2-x peptide is also decreased. Importantly, the loss of meprin beta improved cognitive abilities in APP/lon mice.

Abnormal iron metabolism leads to iron deposition and promotes the progression of Alzheimer's disease. Furthermore, iron deposition can trigger iron-mediated cell death and result in neuronal loss. Therefore, targeting iron metabolism could be a promising therapeutic approach for Alzheimer's disease.

Summary: This study identified MXRA8 as a novel prognostic marker for glioma and highlighted its role in ferroptosis and the immune microenvironment of glioma. Knockdown of MXRA8 inhibited cell viability and increased sensitivity to temozolomide in glioma cells. MXRA8 was also associated with immune infiltration cells and immunoinhibitory molecules. These findings suggest that MXRA8 may serve as a potential therapeutic target for glioma.
