Promising application of a new ulnar nerve compound muscle action potential measurement montage in amyotrophic lateral sclerosis: a prospective cross-sectional study

Summary: Previous studies have shown that the conventional belly-tendon montage does not accurately reflect the action potential of the ulnar nerve. A new method was developed in 2020 to improve the accuracy by adjusting the electrode position. The new method showed more uniform waveforms and may be useful for monitoring amyotrophic lateral sclerosis progression.

Analysis of HTT CAG repeat expansion in Italian patients with amyotrophic lateral sclerosis

Summary: Recent studies have found that full-penetrance pathogenic repeat expansions of the HTT gene exist in a minority of frontotemporal dementia/amyotrophic lateral sclerosis (ALS) patients. In an analysis of ALS patients in Italy, one patient was found to have two expanded alleles of the HTT gene. This patient did not exhibit typical symptoms of Huntington's disease (HD) and had a clinical presentation consistent with ALS, ruling out a misdiagnosis of HD.

The analysis of skin samples from autopsies and biopsies shows a significant increase in alpha Syn(P) seeding activity in individuals with PD and synucleinopathies compared to controls with tauopathies and non-neurodegenerative diseases.

Pathogenic Huntingtin Repeat Expansions in Patients with Frontotemporal Dementia and Amyotrophic Lateral Sclerosis

Summary: Researchers analyzed whole-genome sequence data from 2,442 FTD/ALS patients, 2,599 LBD patients, and 3,158 neurologically healthy subjects, identifying pathogenic HTT gene expansions in a small percentage of FTD/ALS patients but not in the LBD or healthy groups. Postmortem evaluations supported an etiological relationship between HTT repeat expansions and FTD/ALS syndromes, suggesting the importance of genetic screening for HTT repeat expansions in FTD/ALS patients.

A Meta-Analysis of Brain DNA Methylation Across Sex, Age, and Alzheimer's Disease Points for Accelerated Epigenetic Aging in Neurodegeneration

Summary: AD is characterized by specific brain DNA methylation patterns, affected by age and sex. This study analyzed DNAm datasets from healthy adults and AD patients to identify sex-, age-, and AD-associated epigenetic profiles. Results showed shared DNAm differences between males and females across brain regions, with aging affecting cortical regions differently from the cerebellum. AD-related epigenetic modifications were enriched in age-related probes, suggesting accelerated epigenetic aging in the disease.
