SIRT3 regulates mitochondrial biogenesis in aging-related diseases

Summary: SIRT3, the main member of mitochondrial deacetylase family, regulates mitochondrial biogenesis and plays a crucial role in energy metabolism, oxidative stress, and aging. Imbalance of mitochondrial dynamics can impair mitochondrial function and lead to various aging-related diseases. Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC1a) is involved in activating mitochondrial biogenesis through nuclear respiratory factors. This review discusses the involvement of SIRT3 and mitochondrial dynamics in aging-related diseases and highlights the importance of signaling events between SIRT3 and mitochondrial biogenesis in the pathological process of these diseases.

The reduction in dendritic spines of hippocampal CA1 neurons, decreased number of Ki67-positive cells in the hippocampal dentate gyrus and subventricular zone, and impaired proliferation of embryonic neural stem cells suggest that A53T alpha-synuclein may induce adult neurogenesis impairment and cognitive dysfunction.
