Improved Bst DNA Polymerase Variants Derived via a Machine Learning Approach

Synthesis and Polymerase Recognition of Threose Nucleic Acid Triphosphates Equipped with Diverse Chemical Functionalities

Structural insights into TNA synthesis were obtained from a high-resolution X-ray crystal structure analysis, revealing a large cavity in the enzyme active site that accommodates the modification of tUTP substrates. The findings offer a promising route to artificially modified genetic polymers uniformly enhanced with diversity-enhancing functional groups.

Some polymerase mutants can tolerate modifications to synthesize RNA, but challenges must be overcome for efficiently preparing chemically modified RNA.

A biologically stable DNAzyme that efficiently silences gene expression in cells

Summary: Efforts to use RNA-cleaving DNA enzymes as gene-silencing agents have been hindered by low efficacy in clinical trials, but a new xeno-nucleic-acid-modified DNAzyme called X10-23 has shown promise in achieving multiple-turnover activity and resisting nuclease digestion in cellular conditions. X10-23 overcomes product inhibition and facilitates persistent gene silencing in mammalian cells, suggesting new molecular chemotypes can enhance the activity and stability of DNAzymes for potential therapeutic applications.
