Super enhancers as master gene regulators in the pathogenesis of hematologic malignancies

Sonia Debek et al.

Summary: Super enhancers (SE) are long stretches of active enhancers that ensure high expression levels of key genes associated with cell function and survival. Cancer cells remodel their SE landscapes to ensure oncogenic transcriptional reprogramming. SE profiling is a powerful tool for discovering core transcriptional regulatory circuits in tumor cells and provides new insights into cancer cell biology. Targeting SEs opens new therapeutic opportunities for hematologic malignancies.

H3K27ac HiChIP in prostate cell lines identifies risk genes for prostate cancer susceptibility

Claudia Giambartolomei et al.

Summary: The study evaluated the use of chromosome conformation capture combined with immunoprecipitation to identify target genes for prostate cancer GWAS risk loci. The results confirmed previously reported PrCa target genes identified through GWAS/eQTL overlap and identified links between GWAS variants and genes known to play a role in prostate cancer biology. HiChIP was also able to predict enhancer elements at prostate cancer risk loci and confirm their regulatory role in gene expression through CRISPR interference.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Super-enhancers: a new frontier for epigenetic modifiers in cancer chemoresistance

Guo-Hua Li et al.

Summary: Chemoresistance poses a significant challenge in cancer treatment, with super-enhancers (SEs) playing a key role in maintaining cancer cell identity and promoting oncogenic transcription. Inhibition of SE components can decrease oncogenic transcription, inhibit tumor growth, and ultimately achieve combined sensitization and reverse drug resistance effects.

MAP3K8 Is a Prognostic Biomarker and Correlated With Immune Response in Glioma

Jing Ren et al.

Summary: MAP3K8 is overexpressed in glioma and correlates with poor clinicopathological features, serving as an independent prognostic indicator and significantly associated with glioma progression. The gene is mainly enriched in immune-related biological processes, particularly in microglia/macrophage cells of glioma, and positively correlated with immune cell infiltration and immune checkpoint molecules, suggesting its potential role in tumor immunity and as a target for glioma immunotherapy.

Epigenetic mechanisms in breast cancer therapy and resistance

Liliana Garcia-Martinez et al.

Summary: The majority of breast cancers express the estrogen receptor and endocrine therapy has been the mainstay for hormone-responsive breast cancer treatment. Recent studies have shown the importance of the epigenome in determining hormone therapy response and how targeting epigenetic factors can help overcome clinical resistance. Understanding epigenetic mechanisms regulating ER+ breast cancer and endocrine therapy resistance is crucial for improving targeted therapies for this type of cancer.

Landscape of EBV-positive gastric cancer

Motonobu Saito et al.

Summary: Epstein-Barr virus-positive gastric cancer (EBV (+) GC) is a subtype of GC associated with EBV infection, exhibiting unique genomic and clinicopathological features compared to other subtypes. Noncoding RNAs derived from EBV-infected cells play crucial roles in oncogenesis and tumor progression in EBV (+) GC. Recent studies focus on genetic mutations and protein overexpression in EBV (+) GC, proposing potential targeted therapies for this subtype.

ZNF507 affects TGF-β signaling via TGFBR1 and MAP3K8 activation in the progression of prostate cancer to an aggressive state

Wookbong Kwon et al.

Summary: The study identified ZNF507 as a key regulator in the progression of prostate cancer to an aggressive state by upregulating the TGF-beta signaling pathway. ZNF507 was found to promote cell growth, survival, proliferation, and metastasis properties of prostate cancer, and positively regulated the transcription of genes such as TGFBR1, MAP3K8, and FURIN. These findings suggest that ZNF507 could be a promising target for studying the development of advanced metastatic prostate cancers.
