Summary: Pruritus, or itch, is a common symptom associated with various skin and immune disorders. Despite significant progress in understanding the molecular mechanisms behind acute and chronic itch, there are still gaps in our knowledge, making the clinical management of pruritic conditions challenging. Recent research has highlighted the interactions between pruriceptive sensory neurons and different cutaneous cell types, particularly in inflammatory conditions, where these cells release mediators that excite the itch-sensing fibers. There is also bidirectional communication, with sensory neurons releasing mediators that maintain an inflamed and itchy tissue environment.

Atopic dermatitis: Is innate or adaptive immunity in control? A clinical perspective

Summary: Atopic dermatitis is a chronic inflammatory skin disease characterized by barrier defects and immune dysregulations. Epithelial cells, especially keratinocytes, play an important role in the innate immunity of the skin and are involved in the pathogenesis of atopic dermatitis. Filaggrin mutations have been established as an important factor in the development of atopic dermatitis. Current therapeutic approaches focus on targeting cytokines produced by T cells in the adaptive immunity.

A Murine Point Mutation of Sgpl1 Skin Is Enriched With Vγ6 IL17-Producing Cell and Revealed With Hyperpigmentation After Imiquimod Treatment

Summary: This study identified a mouse line carrying a missense mutation of Sgpl1, which successfully modeled a human disease after treatment with Imiquimod. The mutation caused similar pathologies as Sgpl1 knock-out mice in multiple organs but greatly preserved the lifespan. The study also revealed an enrichment of IL17a producing V gamma 6(+) cells in the skin, which contributed to severe skin pathology.
