Plant-based expression and characterization of SARS-CoV-2 virus-like particles presenting a native spike protein

Summary: This study investigated the use of transient expression to produce virus-like particles (VLPs) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of COVID-19, in Nicotiana benthamiana. The native S protein expressed in plants has biological properties similar to those of the parent virus, and the VLPs can be purified by density gradient centrifugation. The study also showed that infiltration with S alone resulted in higher levels of VLPs containing the full-length S protein compared to co-infiltration with M and E proteins. The plant-made material is noninfectious and can be handled without high containment conditions.

Through expressing the RBD of the SARS-CoV-2 spike protein in a glycoengineered tobacco plant, this study successfully generated a candidate subunit vaccine that induced humoral immunity in mice. These findings lay a strong foundation for further development of plant-expressed RBD antigens as effective, safe, and affordable SARS-CoV-2 vaccines, showcasing the potential of plant species in vaccine development.
