In Vitro and In Silico Study of Analogs of Plant Product Plastoquinone to Be Effective in Colorectal Cancer Treatment

Halilibrahim Ciftci et al.

Summary: Plants have provided valuable molecules with diverse biological activities, but sometimes their derivatives need to be developed to enhance the effectiveness. This study evaluated the anticancer effects of PQ2, a derivative of plastoquinone, on colorectal cancer cells. PQ2 demonstrated significant cytotoxicity, induced apoptosis, and showed promising pharmacokinetic properties. It effectively bound to DNA and exhibited unique characteristics for potential use in colorectal cancer treatment.

Future Directions in Chronic Phase CML Treatment

Nathalie Javidi-Sharifi et al.

Summary: This review focuses on recent and emerging treatment paradigms in chronic phase CML, covering new frontline approaches, second treatment-free remission studies, and refractory disease treatment. Specific treatment methods include immunomodulatory approaches, targeting leukemia stem cells, and BCR-ABL1-intrinsic combination therapy. The challenges remain with regard to deepening initial responses, prolonging treatment-free remission, and providing efficacious and tolerable options for patients with refractory disease and resistance mutations.

Design, synthesis and investigation of the mechanism of action underlying anti-leukemic effects of the quinolinequinones as LY83583 analogs

Halil Ciftci et al.

Summary: This study synthesized a series of LY83583 analogs containing alkoxy groups, with AQQ13 showing the most potent cytotoxic activity against K562 CML cell line. AQQ13 induced apoptosis in K562 cells with DNA-cleaving activity, suggesting it as a potential candidate for anti-leukemic drug development.

Structure based design, synthesis, and evaluation of anti-CML activity of the quinolinequinones as LY83583 analogs

Nilufer Bayrak et al.

Summary: Quinone-based small molecules, particularly 6-anilino-5,8-quinolinequinone (LY83583), show promise as antiproliferative drug candidates by inhibiting cyclase and inducing apoptosis in cancer cells. Two series of LY83583 analogs with electron-withdrawing or electron-donating groups were synthesized and evaluated for cytotoxic activity against various cancer cell lines. AQQ15 analog demonstrated high potency and selectivity, inducing apoptotic cell death through DNA cleavage. In silico studies showed AQQ15 to have potential as an anticancer drug candidate with favorable pharmacokinetic parameters.

EGFR in Cancer: Signaling Mechanisms, Drugs, and Acquired Resistance

Mary Luz Uribe et al.

Summary: Growth factors like epidermal growth factor (EGF) can promote cell division and migration, but cancer hijacks these mechanisms for tumor growth and metastasis. Over-production and mutations of EGF and EGFR are common in various cancers, making them popular targets for anti-cancer therapies. The activation of EGFR triggers a series of molecular switches controlling gene expression, with implications for drug resistance and metastasis.

Semi-synthesis as a tool for broadening the health applications of bioactive olive secoiridoids: a critical review

Manuela Oliverio et al.

Summary: This study focuses on the semi-synthetic protocols developed in the last fifteen years for producing natural olive bioactive phenols that are rarely available by extractive procedures, as well as new biophenol derivatives with enhanced biological activity. Additionally, the semi-synthetic protocols for producing olive bioactive phenol derivatives as analytical standards are discussed. The advantages of semi-synthesis compared to classical extraction methods or total synthesis protocols are critically analyzed in this review.
