On the dissociation pathways of copper complexes relevant as PET imaging agents

Rocio Uzal-Varela et al.

Summary: Several bifunctional chelators have been synthesized in recent years for the development of new (64)Cu-based PET agents. High stability and kinetic inertness are important when designing metal-based PET probes. Different competitive assays are commonly used to evaluate the dissociation mechanisms of Cu(II) in the body. This study fills the gap by providing a detailed spectroscopic study of the kinetic behavior of different macrocyclic Cu(II) complexes.

Oriented internal electrostatic fields: an emerging design element in coordination chemistry and catalysis

Alexander B. Weberg et al.

Summary: The power of oriented electrostatic fields (ESFs) to influence chemical bonding and reactivity has attracted growing interest. Strong ESFs have been found to contribute significantly to enzyme activity, where alignment of a substrate's changing dipole moment with a strong, local electrostatic field is responsible for enhancing the enzymatic rate. Researchers have also explored the impacts of internal electrostatic fields through the addition of ionic salts and charged functional groups, as well as externally through bulk fields between electrode plates. Incorporating charged moieties into homogeneous inorganic complexes to generate internal ESFs has great potential for novel catalyst design.

Leveraging slow DOTA f-element complexation kinetics to enable separations by kinetic design

Jessica A. Jackson et al.

Summary: This study explores a new approach using kinetic control in metal ion extraction systems, showing that the rate constant of lanthanide-DOTA complexes increases with the lanthanide series, exhibiting non-monotonic behavior in terbium and thulium. Results suggest that separations between metals are more effective in the early phase of contact, diminishing over longer contact times. The use of slowly complexing ligands like DOTA may be suitable for kinetically based systems, providing a new design basis for f-element separations.

Copper Coordination Chemistry of Sulfur Pendant Cyclen Derivatives: An Attempt to Hinder the Reductive-Induced Demetalation in 64/67Cu Radiopharmaceuticals

Marianna Tosato et al.

Summary: A series of Cu2+ complexes formed by cyclen derivatives with sulfur pendant arms were studied for their potential as chelators for copper radioisotopes. Various experiments were conducted to evaluate the stoichiometry and stability of these complexes, demonstrating their promising application in copper-based radiopharmaceuticals. The high stability of both Cu2+ and Cu+ complexes suggests these sulfur-containing chelators may prevent in vivo demetalation after bioinduced reduction to Cu+.
