Ruthenium(II) Polypyridyl Complexes and Their Use as Probes and Photoreactive Agents for G-quadruplexes Labelling

Julie Jiang et al.

Summary: Ruthenium(II) polypyridyl complexes have been widely used in various applications due to their optical and electrochemical properties. The use of photoprobes to mark and understand the hidden characteristics of G-quadruplexes (G4s) has recently garnered attention. This review provides an overview of the ligands and metal complexes that interact with G4s, as well as a comprehensive summary of the interactions and (photo)reactions of Ru(II) complexes with G4s.

Photo-induced telomeric DNA damage in human cancer cells

Justin Weynand et al.

Summary: We report on the study of novel dinuclear ruthenium(ii) complexes designed to target and react with G-quadruplex telomeric DNA. These complexes efficiently generate guanine radical cation sites as photo-oxidation products upon irradiation, and show strong photocytotoxicity towards osteosarcoma cells. Microscopic-based telomere dysfunction assay provides direct evidence of forming photo-oxidative damage at telomeres in cells, highlighting the potential for future cancer phototherapies.

BrdU immuno-tagged G-quadruplex ligands: a new ligand-guided immunofluorescence approach for tracking G-quadruplexes in cells

Thibaut Masson et al.

Summary: This study presents a novel visualization strategy based on the immuno-recognition of 5-bromo-2'-deoxyuridine (5-BrdU) modified G4 ligands, functionalized before or after binding to G4 targets by CuAAC. The recognition of the tag by antibodies allows for the exclusive detection of the modified ligands when bound to a G4 target both in vitro and in cells, leading to a highly efficient G4-ligand Guided Immunofluorescence Staining (G4-GIS) approach.

The Cellular Functions and Molecular Mechanisms of G-Quadruplex Unwinding Helicases in Humans

Yang Liu et al.

Summary: G-quadruplexes (G4s) are stable non-canonical secondary structures formed by G-rich DNA or RNA sequences, playing important regulatory roles in biological processes. G4 unwinding helicases are key players in G4 metabolism and function, closely linked to physiological and pathological processes. By systematically summarizing the functions and molecular mechanisms of human G4 unwinding helicases, this review provides researchers with opportunities to explore the frontiers in the functions and molecular mechanisms of human G4 unwinding helicases.

G-quadruplex stabilization via small-molecules as a potential anti-cancer strategy

Annoor Awadasseid et al.

Summary: G-quadruplexes, secondary DNA structures consisting of guanine-rich nucleic acids, play a crucial role in cancer therapy by stabilizing secondary DNA structures in oncogenes' promoters. Targeting specific protein products of genes with abnormal expression poses challenges due to structural difficulties, protein overexpression, and mutations affecting treatment resistance, thus the use of small molecules to stabilize these DNA structures presents a new strategy.

Identifying G-Quadruplex-DNA-Disrupting Small Molecules

Jeremie Mitteaux et al.

Summary: This article reports on efforts to develop in vitro assays to reliably identify molecules that can destabilize G4s. The workflow includes a newly designed G4-unfold assay, adapted from the G4-helicase assay with Pif1, as well as biophysical and biochemical techniques traditionally used to study G4/ligand interactions, and a qPCR stop assay adapted from a Taq-based protocol to identify G4s in genomic DNA. This multipronged approach leads to the characterization of a phenylpyrrolocytosine (PhpC)-based G-clamp analog as a prototype of a G4-disrupting small molecule.

DNA G-quadruplex structures: more than simple roadblocks to transcription?

Jenna Robinson et al.

Summary: The relationship between G-quadruplex (G4) secondary structures in gene promoters and gene expression regulation has been studied for over 20 years. While originally thought to act as transcriptional repressors, recent research has challenged this simplistic model by suggesting that G4 formation can alter gene expression at various levels, serving as key regulatory elements disrupting epigenetic marks and chromatin architecture.
