Single particle cryo-EM structure of the outer hair cell motor protein prestin

Carmen Butan et al.

Summary: This study reports the cryo-EM structure and characterization of gerbil prestin, a protein that drives OHC electromotility. Prestin is structurally similar to Slc26a9 and is captured in an inward-open state. The study also reveals the effect of mutation in prestin's chloride binding site on its function.

Single-cell transcriptome analysis reveals three sequential phases of gene expression during zebrafish sensory hair cell regeneration

Sungmin Baek et al.

Summary: In this study, the transcriptional dynamics of sensory hair cell regeneration in zebrafish were characterized using single-cell RNA sequencing. Three sequentially activated modules were uncovered, providing insights into the gene regulatory networks involved in hair cell regeneration. These findings are important for understanding hair cell regeneration and triggering regeneration in mammals.

Modeling of mitochondrial bioenergetics and autophagy impairment in MELAS-mutant iPSC-derived retinal pigment epithelial cells

Sujoy Bhattacharya et al.

Summary: This study utilized MELAS iPSC-derived RPE cells to demonstrate how mitochondrial DNA alterations influence RPE function, particularly in aging and macular degeneration. The research found that MELAS iPSC-derived RPE cells exhibited various characteristics, including reliance on glycolysis for energy generation and impaired autophagy.

Mitochondrial Cristae Architecture and Functions: Lessons from Minimal Model Systems

Frederic Joubert et al.

Summary: Mitochondria are considered as the powerhouses of eukaryotic cells, with energy production occurring in specific dynamic membrane invaginations. The integrity of these structures is crucial for proper mitochondrial function, yet mechanisms behind their plasticity and organization, and their influence on mitochondrial function remain less understood. This review focuses on studies questioning the role of lipid membrane composition using minimal model systems.

Building inner ears: recent advances and future challenges for in vitro organoid systems

Wouter H. van der Valk et al.

Summary: While inner ear disorders are common, our ability to intervene and recover their sensory function is limited. Developing in vitro models such as inner ear organoids could aid in identifying new regenerative drugs and gene therapies. To enhance the applicability of in vitro inner ear models, a more holistic view focusing on the broader diversity of neuroglial and mesenchymal cell types is needed.

Phase-Locking Requires Efficient Ca2+ Extrusion at the Auditory Hair Cell Ribbon Synapse

Adolfo E. Cuadra et al.

Summary: The study found that clearance of Ca2+ dynamics in paired hair cell synapses follows a double exponential function, with the fast component significantly reduced under prolonged stimulation. Inhibiting PMCA, MCU, or SERCA increased both basal Ca2+ levels and the Ca2+ rise during sinusoidal stimulation, while significantly reducing phase-locking of synaptic vesicle releases from hair cells.

Using the Zebrafish Lateral Line to Understand the Roles of Mitochondria in Sensorineural Hearing Loss

Melanie Holmgren et al.

Summary: Hair cells in the inner ear play a crucial role in hearing, but can be damaged by various factors. Mitochondria in hair cells are key in producing energy and maintaining calcium balance, and their dysfunction can lead to hearing loss. Zebrafish provide a valuable model to study hair-cell mitochondrial activity and develop therapeutic strategies for sensorineural hearing loss.

Synaptic Release Potentiation at Aging Auditory Ribbon Synapses

Thibault Peineau et al.

Summary: Age-related hidden hearing loss is often caused by a progressive degeneration of inner hair cell (IHC) ribbon synapses. This study found that a decrease in synapse number may lead to hearing loss and potentially cause hyperacusis. Synaptic transmission at surviving synapses was found to be functionally potentiated, partially compensating for the decrease in synapse number.

Postnatal structural development of mammalian Basilar Membrane provides anatomical basis for the maturation of tonotopic maps and frequency tuning

Tomomi Tani et al.

Summary: Using polarized light microscopy, this study characterized the architectural transformations of the basilar membrane (BM) of the mammalian cochlea during postnatal development and maturation. The results indicate that the structural elements of the BM increase progressively in size and become more tightly aligned with maturation, reaching the adult structural signature by postnatal day 20. The findings provide insight into the structural details and developmental changes of the mammalian BM, suggesting that it is a dynamic structure that changes throughout an animal's life.

Mitochondrial calcium uniporter is essential for hearing and hair cell preservation in congenic FVB/NJ mice

Mayakannan Manikandan et al.

Summary: Mitochondrial Ca2+ uniporter is crucial for maintaining Ca2+ balance in hair cells, and plays a key role in hearing function. Knockout of MCU in mice leads to hearing impairment and loss of hair cells, but does not affect cochlear development.
