Summary: This study investigates the pathological mechanisms of migraine chronification by measuring the levels of GABA and Glx in the DN and PAG of migraine patients and healthy controls. The results suggest that chronic migraine patients have lower GABA levels in the DN and higher Glx levels in the PAG compared to healthy controls. Higher GABA levels in the PAG are associated with poorer sleep quality in all migraine patients, while lower Glx levels in the DN may be associated with more severe migraine disability. This study provides evidence of neurochemical alterations in the DN and PAG in migraine chronification and sheds light on the pathological mechanisms underlying migraine characteristics.
