Chromoanagenesis, the mechanisms of a genomic chaos

Summary: Designated as chromoanagenesis, the phenomena of chromothripsis, chromanasynthesis, and chromoplexy are new types of complex rearrangements that have changed our understanding of chromosomal abnormalities. These rearrangements typically occur within a single cell cycle and are closely linked to genomic instability. The formation of micronuclei integrating isolated chromosomal material and the occurrence of chromatin bridges resulting from telomeric fusions are two major mechanisms that generate chromoanagenesis. These mechanisms are consistent with the features of complex chromosomal rearrangements associated with chromoanagenesis.

Summary: This report discusses a 14-year-old boy, his father, and paternal uncle, all carriers of a duplication of a chromosomal region. The duplication was transmitted by the grandmother and involved more than 40 genes without apparent clinical effects through two generations. Whole genome sequencing was performed on the boy's father to determine the molecular basis of this structural variant.

Deciphering complex genome rearrangements in C. elegans using short-read whole genome sequencing
