Interaction-based Mendelian randomization with measured and unmeasured gene-by-covariate interactions

Summary: Studies on gene-environment interactions in Mendelian randomization have led to the development of two similar methodologies: MR-GxE and MR-GENIUS. These methods correct for pleiotropic bias when using individual instruments. We critically examine the assumptions of both methods and propose sensitivity analyses to evaluate their performance. Using data from the UK Biobank, we investigate the effect of BMI on SBP and find evidence of a positive effect. While both approaches share similar assumptions, their differences make them suitable for different research settings.
