Dihydroquinazolin-4(1H)-one derivatives as novel and potential leads for diabetic management

Summary: A series of dihydroquinazolin-4(1H)-one derivatives were synthesized in this study and evaluated for their inhibitory activities against alpha-amylase and alpha-glucosidase enzymes. The compounds showed significant to moderate inhibition potential, with different substituents affecting their inhibitory potency. Kinetic studies revealed competitive inhibition for alpha-amylase and non-competitive inhibition for alpha-glucosidase, while in silico analysis identified important binding interactions with the active sites of the enzymes.

Isatin thiazoles as antidiabetic: Synthesis, in vitro enzyme inhibitory activities, kinetics, and in silico studies

Evaluation of synthetic 2-aryl quinoxaline derivatives as α-amylase, α-glucosidase, acetylcholinesterase, and butyrylcholinesterase inhibitors

Summary: A variety of 2-aryl quinoxaline derivatives were successfully synthesized in good yields by reacting 1,2-phenylenediamine with different substituted phenacyl bromides in the presence of pyridine catalyst. These synthetic derivatives showed enhanced inhibitory potential against alpha-amylase, alpha-glucosidase, acetylcholinesterase, and butyrylcholinesterase at nanomolar concentrations, with compound 14 demonstrating superior activity compared to standard inhibitors. Selected potent compounds were further analyzed through molecular docking studies to understand their binding energies and interactions with the target enzymes.

Biology-oriented drug synthesis of nitrofurazone derivatives: Their α-glucosidase inhibitory activity and molecular docking studies

Summary: In this study, twenty structural variants of nitrofurazone were synthesized using a biology-oriented drug synthesis approach. The synthesized molecules were characterized using spectroscopic techniques, and their alpha-glucosidase inhibitory activity was determined. Several compounds showed potent inhibition against alpha-glucosidase, with compound 19 being the most active. Molecular docking and dynamic simulation were used to study the binding interactions and stability of the active compounds.

Dicyanoanilines as potential and dual inhibitors of α-amylase and α-glucosidase enzymes: Synthesis, characterization, in vitro, in silico, and kinetics studies

Summary: The study focuses on the synthesis of dicyanoaniline derivatives and their inhibition activities against alpha-amylase and alpha-glucosidase enzymes. Among the synthesized compounds, five of them showed excellent inhibition against the enzymes. Enzyme kinetic studies revealed different types of inhibition mechanisms, and computer simulations demonstrated the involvement of these molecules in binding interactions at the enzyme's active site.

Summary: The cattle tick, Rhipicephalus microplus, is the most damaging pest in tropical and sub-tropical areas of the world. This study aimed to develop phytochemical-based acaricides from Berberium lyceum and Tamarixa aphylla to control Rhipicephalus microplus populations. The results showed that the bioactive components from these plants exhibited significant oviposition and larval mortality inhibition.

A facile approach synthesis of benzoylaryl benzimidazole as potential α-amylase and α-glucosidase inhibitor with antioxidant activity

Summary: A series of benzoylarylbenzimidazole compounds were synthesized from specific routes and evaluated for their antioxidant and antidiabetic enzyme activities, showing significant activity for most compounds. Compound 3f exhibited remarkable inhibition against alpha-amylase and alpha-glucosidase, comparable to the reference drug acarbose. In silico molecular docking calculations confirmed the strong binding affinity of compounds 3a-h with human pancreatic alpha-amylase and human lysosomal acid-alpha-glucosidase active sites, supporting the excellent activity of most tested compounds.
