Rapid directed molecular evolution of fluorescent proteins in mammalian cells

Siranush Babakhanova et al.

Summary: This study presents a practical method for rapid optimization of fluorescent proteins in cultured mammalian cells via directed molecular evolution, resulting in a set of green and near-infrared fluorescent proteins with enhanced intracellular brightness. These proteins exhibit high performance in fluorescent labeling of neurons in culture and in vivo in small model organisms, allowing for crosstalk-free multicolor imaging. This approach has the potential to be widely adopted by protein engineers due to its simplicity and practicality.

Summary: This study presents the first systematic investigation of in vivo maturation of 12 fluorescent proteins in budding yeast, along with a new method for accurately estimating maturation rates. The approach overcomes technical limitations and can be applied to a wide range of organisms, providing a useful resource for the research community.

Overexpression of SARS-CoV-2 protein ORF6 dislocates RAE1 and NUP98 from the nuclear pore complex

Summary: Studies have shown that the ORF6 protein of SARS-CoV-2 can manipulate the localization and functions of nucleoporins, leading to abnormal nucleocytoplasmic trafficking and ultimately reducing the size of host cell nucleus and halting cell growth.
