Mitochondrial Ferritin Deficiency Promotes Osteoblastic Ferroptosis Via Mitophagy in Type 2 Diabetic Osteoporosis

XinDong Wang et al.

Summary: The incidence of type 2 diabetic osteoporosis (T2DOP) is rapidly increasing and its specific mechanism remains unclear. Studies have shown a relationship between iron overload and T2DOP. Mitochondrial ferritin (FtMt) may inhibit ferroptosis in osteoblasts by reducing oxidative stress caused by excess ferrous ions, while FtMt deficiency induces mitophagy in the pathogenesis of T2DOP. FtMt could potentially serve as a target for T2DOP therapy.

CD8+ T cells and fatty acids orchestrate tumor ferroptosis and immunity via ACSL4

Peng Liao et al.

Summary: T cell-derived interferon gamma (IFN γ) in combination with arachidonic acid (AA) induces immunogenic tumor ferroptosis, serving as a mechanism for CD8(+) T cell-mediated tumor killing. This process involves IFN γ stimulation of ACSL4 and alteration of tumor cell lipid pattern.

Bortezomib limits renal allograft interstitial fibrosis by inhibiting NF-κB/TNF-α/Akt/mTOR/P70S6K/Smurf2 pathway via IκBα protein stabilization

Chuanjian Suo et al.

Summary: The study showed that bortezomib attenuated TNF-α-induced epithelial-mesenchymal transition and renal allograft interstitial fibrosis by inhibiting NF-κB/TNF-α/Akt/mTOR/p70S6 kinase/Smurf2 signaling and stabilizing the inhibitor of NF-κB protein.

Ferroptosis: mechanisms, biology and role in disease

Xuejun Jiang et al.

Summary: Ferroptosis, as a form of regulated cell death driven by iron-dependent phospholipid peroxidation, has seen significant growth in research in recent years. Studies have focused on molecular mechanisms, regulation, and functions of ferroptosis, linking this cell death modality to various pathologies and proposing its roles in normal physiology and potential therapeutic targeting.

Legumain promotes tubular ferroptosis by facilitating chaperone-mediated autophagy of GPX4 in AKI

Chuan'ai Chen et al.

Summary: The study demonstrates that deficiency of legumain attenuates acute renal tubular injury, inflammation, and ferroptosis in acute kidney injury models induced by ischemia-reperfusion injury or folic acid. Legumain deficiency prevents chaperone-mediated autophagy of GPX4, dampening ferroptosis induced by hypoxia or erastin. The results suggest that legumain promotes tubular ferroptosis in acute kidney injury by promoting chaperone-mediated autophagy of GPX4.

Chronic Allograft Injury

Eric Langewisch et al.

Summary: With the improved long-term kidney transplant survival, attention has shifted to understanding the causes of transplanted allograft failure. Chronic allograft injury involves both immune-mediated and nonimmune-mediated injuries, presenting an opportunity to identify those at greater risk and implement new strategies. The research highlights the need to identify markers of injury and mechanisms of injury in order to better understand late graft failure.

Impaired ATG16L-Dependent Autophagy Promotes Renal Interstitial Fibrosis in Chronic Renal Graft Dysfunction Through Inducing EndMT by NF-κB Signal Pathway

Zeping Gui et al.

Summary: The study identified that low expression of ATG16L in patients with chronic renal graft dysfunction (CAD) compared to non-CAD patients, and suppressed autophagic flux is a poor prognostic factor. Knockdown of ATG16L enhanced EndMT process in human renal glomerular endothelial cells (HRGECs). In rat renal transplant models of CAD, the abundance of ATG16L was accompanied by dynamic changes in autophagic flux during different stages of kidney transplantation.

A Higher Foci Density of Interstitial Fibrosis and Tubular Atrophy Predicts Progressive CKD after a Radical Nephrectomy for Tumor

Luisa Ricaurte Archila et al.

Summary: IF/TA density (the number of IF/TA foci per area of cortex) predicts progressive CKD and noncancer mortality after nephrectomy, independent of the percentage of IF/TA. A higher density of IF/TA foci suggests more scattered damage to the nephrons and worse kidney prognosis at the same percentage of IF/TA.

Liproxstatin-1 attenuates unilateral ureteral obstruction-induced renal fibrosis by inhibiting renal tubular epithelial cells ferroptosis

Bo Zhang et al.

Summary: The study demonstrates the role of ferroptosis in renal fibrosis induced by unilateral ureteral obstruction. Inhibiting ferroptosis can alleviate the morphological and pathological changes of renal fibrosis and suppress the expression of profibrotic factors. Liproxstatin-1 may potentially be used as a therapeutic approach for UUO-induced renal fibrosis by inhibiting ferroptosis in renal tubular epithelial cells.

Summary: Our research demonstrated the involvement of ferroptosis in the development of DN, and up-regulating Nrf2 with fenofibrate treatment inhibited diabetes-related ferroptosis, delaying the progression of DN. This study provided a new perspective on the development mechanism of DN and offered a new approach to delay the progression of DN.
