Summary: Decidual macrophages (dMφ), important regulators of maternal-fetal immune tolerance and trophoblast invasion during pregnancy, display an active glycerophospholipid metabolism and their adhesion and retention, as well as M2 differentiation, are facilitated by the activation of ENPP2-lysophosphatidic acid (LPA). This process involves the activation of LPA receptors, mediated through the DDIT4-macroautophagy/autophagy axis, leading to upregulation of multiple adhesion factors and poor trophoblast invasion. Supplementation with LPA or autophagy inducer rapamycin improves dMφ autophagy and cell residence, preventing spontaneous abortion.
