A Biomimetic Human Lung-on-a-Chip with Colorful Display of Microphysiological Breath

Yujuan Zhu et al.

Summary: In this study, a novel biomimetic 3D microphysiological lung-on-a-chip system with breathing visualization was developed. The system, inspired by the iridescence phenomenon of soap bubbles, allows for the cyclic deformation of pulmonary alveoli and real-time monitoring of cell mechanics. The research also reveals the essential role of mechanical stretching in idiopathic pulmonary fibrosis.

In vitro and ex vivo models in inhalation biopharmaceutical research - advances, challenges and future perspectives

Mohammed Ali Selo et al.

Summary: Oral inhalation is the preferred drug delivery method for respiratory disorders, reducing systemic side effects. The high alveolar surface area, low enzymatic activity, and rich blood supply of distal airspaces offer a promising pathway to systemic circulation. Various cell and tissue-based models have been developed to mimic pulmonary biological barriers and address biopharmaceutical questions.

A Biomimetic, Copolymeric Membrane for Cell-Stretch Experiments with Pulmonary Epithelial Cells at the Air-Liquid Interface

Ali Doryab et al.

Summary: The study presents a bioinspired lung membrane that can mimic the complex environment of the lung, with good stretchability and surface properties for culturing lung epithelial cells. Physiological cyclic stretch improves cell structure and function, while non-physiological over-stretch leads to cell death and inflammatory response.

Optimization of an air-liquid interface in vitro cell co-culture model to estimate the hazard of aerosol exposures

Rui-Wen He et al.

Summary: Inhalation exposure to aerosol contaminants can lead to respiratory health problems. To mimic long-term exposure for toxicity testing, lung epithelial cells must be maintained under ALI conditions and co-cultured with macrophages. Calu-3 cells maintain monolayer structure and tight junctions, making them ideal for co-culturing with macrophages for increased sensitivity in responses to aerosol exposure.

Oliver Brookes et al.

Summary: Research on the development of an air-liquid interface model of alveolar epithelium incorporating features of type I and type II epithelial cells showed promising results in terms of cell morphology and barrier function. The coculture model demonstrated stable barrier properties similar to native alveolar epithelium, indicating its potential for representing a broader range of functions.
