Dithiocarbamates effectively inhibit the α-carbonic anhydrase from Neisseria gonorrhoeae

Summary: In this study, a class of compounds called dithiocarbamates (DTCs) was found to efficiently inhibit the carbonic anhydrase enzyme in Neisseria gonorrhoeae, with better inhibition than other inorganic anions and sulphonamides. Three of these compounds showed low minimum inhibitory concentration values in antimicrobial testing, suggesting their potential as novel antigonococcal drugs.

Exploring the structure-activity relationships of diphenylurea as an antibacterial scaffold active against methicillin- and vancomycin- resistant Staphylococcus aureus

Summary: A series of diphenylurea derivatives with aminoguanidine moiety were synthesized and their antibacterial activity was evaluated. Two compounds showed better bacteriological profiles and prolonged post-antibiotic effect. Additionally, these compounds were able to resensitize vancomycin-resistant strains and exhibited comparable efficacy to a commercially available drug in a mouse model. Therefore, the diphenylurea scaffold has potential as an anti-staphylococcal treatment option.

Isoquinoline Antimicrobial Agent: Activity against Intracellular Bacteria and Effect on Global Bacterial Proteome

Summary: This study presents a new class of alkynyl isoquinoline antibacterial compounds, synthesized via Sonogashira coupling, which exhibit strong bactericidal activity against a range of Gram-positive bacteria including methicillin- and vancomycin-resistant Staphylococcus aureus strains. Representative compounds in this class, HSN584 and HSN739, are capable of reducing methicillin-resistant S. aureus load in macrophages and show a low propensity for developing resistance. Comparative global proteomics and macromolecule biosynthesis assays suggest that HSN584 perturbs S. aureus cell wall and nucleic acid biosynthesis. The alkynyl isoquinoline moiety holds promise as a new scaffold for the development of potent antibacterial agents against multidrug-resistant Gram-positive bacteria.
