Design, synthesis, and biological evaluation of novel morpholinated isatin-quinoline hybrids as potent anti-breast cancer agents

Atamjit Singh et al.

Summary: Novel hydrazine-linked morpholinated isatin-quinoline hybrids were designed, synthesized, and evaluated as potent and safer anti-breast cancer agents, showing high inhibitory potential against hormone-positive breast cancer cells with selective action and mitotic arrest at the G(2)/M phase, suggesting AS-4 as a potential lead compound for further development.

Design, synthesis, and molecular modeling of quinoline-based derivatives as anti-breast cancer agents targeting EGFR/AKT signaling pathway

Rasha Z. Batran et al.

Summary: Two series of quinoline-thiazole and quinoline-thiazolidinone hybrids were synthesized and evaluated for their in vitro antitumor activity. Compound 4b and 6b showed better antiproliferative activity than lapatinib, with compound 4b also exhibiting better inhibitory activity towards the EGFR pathway. Compound 4b was capable of down-regulating anti-apoptotic genes and up-regulating a pro-apoptotic gene.

Crystal structure of the ternary complex of Leishmania major pteridine reductase 1 with the cofactor NADP+/NADPH and the substrate folic acid

Lucia Dello Iacono et al.

Summary: This study presents the high-resolution crystal structure of LmPTR1 in complex with folic acid and describes the intermolecular forces that drive substrate binding. By expanding the collection of LmPTR1 structures in complex with process intermediates, additional insights into the active-site rearrangements during catalysis are provided. A small but significant difference in the orientation of the catalytic triad was found compared to previous structures.

Recent update on antibacterial and antifungal activity of quinoline scaffolds

Atukuri Dorababu

Summary: Although most heterocycles have significant pharmacological activities, only a few quinoline derivatives exhibit the best biological activities, including anticancer, anti-inflammatory, antibacterial, antiviral, and antifungal activities. Therefore, designing more efficient antimicrobial drugs is a priority to prevent malevolent microbial diseases.

Novel oxindole/benzofuran hybrids as potential dual CDK2/GSK-3β inhibitors targeting breast cancer: design, synthesis, biological evaluation, and in silico studies

Wagdy M. Eldehna et al.

Summary: In this study, three series of oxindole-benzofuran hybrids were designed and synthesised as dual CDK2/GSK-3 beta inhibitors targeting breast cancer. The N-1 -unsubstituted oxindole derivatives showed potent cytotoxic activity, while the N-1-substituted derivatives exhibited moderate to weak activity on breast cancer cell lines. The most potent compounds caused cell cycle arrest in the G2/M phase by inhibiting CDK2/GSK-3 beta, as demonstrated by enzyme inhibition assays and molecular docking studies.

Yousry A. Ammar et al.

Summary: The study involved synthesis of quinoline-based thiazole analogues for antibiotic class, which showed good antimicrobial activity against standard microbial pathogens and MDR bacterial strains. Active derivatives were effective in treating bacterial infection by inhibiting DHFR enzyme, with molecular docking study supporting the binding mode. DFT calculation generated MEP surfaces to determine interaction regions for different types of interactions.
