Design and synthesis of new quinoline derivatives as selective C-RAF kinase inhibitors with potent anticancer activity

Summary: This article presents the design, synthesis, and biological screening results of a new series of diarylurea and diarylamide derivatives with a quinoline core armed with dimethylamino or morpholino side chains. The compounds showed broad-spectrum antiproliferative activity against a panel of 60 cancer cell lines, with three of them demonstrating higher potency than the reference drug, sorafenib. The compounds also exhibited high selectivity for C-RAF kinase, making them potential inhibitors for this molecular target.

Carbonic anhydrase IX: a regulator of pH and participant in carcinogenesis

Summary: Carbonic anhydrase IX (CAIX) is a transmembrane metalloenzyme upregulated in tumor cells under hypoxic conditions. High expression of CAIX is associated with poor prognosis in breast, lung, ovarian, and bladder carcinomas, while low expression indicates poor prognosis in clear cell renal cell carcinoma (CCRCC). CAIX immunohistochemistry is useful for diagnosing metastatic CCRCC and clear cell papillary renal cell carcinoma, and there is interest in targeting it with monoclonal antibodies and small molecule inhibitors.
