Summary: This study found that the gene ENPP1, which causes abnormal mineralization, is associated with AFF. This suggests that osteomalacia is an underlying condition predisposing to AFF and that higher incident rates of AFFs in Asian populations might be explained by the genetic risk factors including ENPP1.

Gene Network of Susceptibility to Atypical Femoral Fractures Related to Bisphosphonate Treatment

Summary: Atypical femoral fractures (AFF) are rare fragility fractures in the subtrochanteric or diaphysis femoral region associated with long-term bisphosphonate (BP) treatment. Genetic analysis identified multiple genes, including DAAM2 and LRP5, as being associated with AFF. Gene interaction analysis revealed a complex network among bone-related genes and other mutated genes. Biological function analysis highlighted cytoskeleton and cilium organization as potentially relevant to AFF.

Summary: Atypical femur fractures (AFFs) are rare complications of anti-resorptive therapy. This study identified rare genetic variants associated with AFF in two Singaporean families. Although the results are inconclusive, the familial aggregation of AFFs suggests a genetic component in AFF pathogenesis.

Summary: Mutations in CYP24A1 and SLC34A3 genes were found in patients with hypercalcemia, with most cases associated with renal diseases. Some patients had hypophosphatemia due to CYP24A1 mutations, while others had normal phosphatemia with SLC34A3 mutations. Variations in SLC34A1 and SLC34A3 showed uncertain significance, and some heterozygous relatives may present with biochemical changes in mineral metabolism.
