Growth Hormone and Insulin-Like Growth Factor 1 Regulation of Nonalcoholic Fatty Liver Disease

Laura E. Dichtel et al.

Summary: This article summarizes the high prevalence of nonalcoholic fatty liver disease (NAFLD) in patients with obesity and its progression to nonalcoholic steatohepatitis (NASH). Understanding the etiology of NAFLD is clinically relevant as it is an independent risk factor for diabetes and cardiovascular disease. The potential role of growth hormone and insulin-like growth factor 1 in regulating hepatic steatosis, inflammation, and fibrosis is discussed.

Reversible insulin resistance in muscle and fat unrelated to the metabolic syndrome in patients with acromegaly

Mai C. Arlien-Soborg et al.

Summary: This study investigates the molecular mechanisms of insulin resistance dissociated from obesity in patients with acromegaly. The results suggest that uncontrolled lipolysis is a major feature of insulin resistance in active acromegaly, characterized by upregulation of PTEN and suppression of insulin signaling in both muscle and fat.

Expression of IGF-1 receptor and GH receptor in hepatic tissue of patients with nonalcoholic fatty liver disease and nonalcoholic steatohepatitis

Stephanie A. Osganian et al.

Summary: This study investigated the expression of IGF-1R and GHR in the livers of NAFLD patients using gene expression analysis and immunohistochemistry. The results showed that there were no significant differences in the expression levels of IGF-1R and GHR across NAFLD severity, while the expression of IGF-1 decreased with increasing severity of NAFLD.

The GH/IGF-1 Axis Is Associated With Intrahepatic Lipid Content and Hepatocellular Damage in Overweight/Obesity

Laura E. Dichtel et al.

Summary: Individuals with NAFLD have lower peak-stimulated GH levels but similar IGF-1 levels compared to controls. Higher peak-stimulated GH levels are associated with lower intrahepatic lipid content (IHL) and less hepatocellular damage. Higher IGF-1 levels are associated with more favorable fibrosis risk scores.

Summary: The study found that knocking down GHR in adult hepatocytes leads to non-alcoholic fatty liver disease, with no significant changes observed in female mice but with symptoms of steatosis, hepatocyte ballooning, and inflammation present in male mice.
