Elucidating Sequence and Structural Determinants of Carbohydrate Esterases for Complete Deacetylation of Substituted Xylans

Summary: Acetylated glucuronoxylan is a common type of hemicellulose, and the acetyl xylan esterase (AcXE) from Flavobacterium johnsoniae is the first enzyme that can remove specific acetyl groups from the glucuronoxylan structure. In this study, the crystal structures of six homologs of FjoAcXE were solved, providing insights into the key structural features of this enzyme family.

Summary: Lignin-carbohydrate complexes (LCCs) play a significant role in the decomposition of glucuronoxylans, and the addition of different enzymes can promote the release of MeGlcpA, with a certain proportion of it being occupied by LCCs. Insoluble LCC fractions significantly reduce the concentration of soluble alpha-glucuronidase, while the reduction in soluble TtCE15A is less pronounced.
