NANOG prion-like assembly mediates DNA bridging to facilitate chromatin reorganization and activation of pluripotency

Kyoung-Jae Choi et al.

Summary: Choi et al. found that human NANOG proteins can form gel-like condensates at low concentrations, which play a crucial role in maintaining pluripotency. The unique features of NANOG, including its largely disordered structure and prion-like domain, contribute to its dose-sensitive function as a master transcription factor. NANOG oligomerization is essential for bridging DNA elements and specific chromatin interactions.

Dilute phase oligomerization can oppose phase separation and modulate material properties of a ribonucleoprotein condensate

Ian Seim et al.

Summary: This study reveals a mechanism that influences the composition of ribonucleoprotein bodies, in which specific sequence motifs and synergies with the intrinsically disordered region drive the formation of oligomers in the dilute phase, hindering its incorporation into the dense phase. Phase-separating RNA-binding proteins form heterogeneous distributions of clusters in subsaturated solutions

Mrityunjoy Kar et al.

Summary: Macromolecular phase separation is an important process in the formation of membraneless biomolecular condensates in cells. In this study, the presence of heterogeneous distributions of clusters in subsaturated solutions of phase-separating RNA-binding proteins was observed, contradicting classical nucleation theory. The size of clusters increased as protein concentrations approached saturation, with some clusters containing tens to hundreds of molecules. Cluster formation in subsaturated solutions and phase separation in supersaturated solutions were found to be strongly coupled, and could be decoupled using solutes and specific mutations.

Mutations linked to neurological disease enhance self-association of low-complexity protein sequences

Xiaoming Zhou et al.

Summary: Protein domains of low sequence complexity do not form stable, three-dimensional structures. However, they play a role in assisting cell organization by self-associating through labile structures. Studies have shown that the interactions responsible for this self-association can be influenced by the equilibrium between folded and unfolded states of the protein structures. Furthermore, certain missense mutations enhance the stability of the labile structures formed during LCD self-association, leading to the manifestation of diseases.

Mechanistic Models of Protein Aggregation Across Length-Scales and Time-Scales: From the Test Tube to Neurodegenerative Disease

Georg Meisl et al.

Summary: Advances in the understanding of protein aggregation in neurodegenerative diseases have been made, but the molecular mechanisms underlying aggregate proliferation in affected individuals are still poorly understood. In vitro studies have shed light on the assembly mechanisms of purified protein molecules, but applying these findings to disease-relevant systems presents challenges. Modifying experimental approaches and analysis techniques for in vivo studies is necessary to gain a better understanding of the mechanisms of protein aggregation.

Summary: The concentrations of macromolecules within cells can affect the conformations and dynamics of proteins. By reducing cell volume through hyperosmotic stress, it was found that intrinsically disordered proteins exhibit compaction and slower dynamics in more crowded cells. Summary: Liquid-liquid phase separation (LLPS) is a unique means of forming biomolecular condensates in organisms, playing significant roles in various biological processes. Post-translational modifications (PTMs), including phosphorylation, methylation, citrullination, acetylation, ubiquitination and poly(ADP-ribosyl)ation, are important regulators of LLPS, affecting protein properties and phase transition behavior. Understanding the regulatory mechanisms of LLPS, especially the roles of PTMs, RNA and molecular chaperones, is essential for potential therapeutic interventions in diseases related to abnormal phase transitions.
