The assembly, regulation and function of the mitochondrial respiratory chain

Irene Vercellino et al.

Summary: The mitochondrial oxidative phosphorylation system is essential for cellular metabolism, driving ATP production through the electron transport chain. Recent studies have provided insights into the organization, assembly, and mechanisms of the respiratory complexes, including the formation of larger assemblies such as respiratory supercomplexes, and their roles in physiology.

GWAS and ExWAS of blood mitochondrial DNA copy number identifies 71 loci and highlights a potential causal role in dementia

Michael Chong et al.

Summary: Mitochondrial DNA concentration in blood samples is associated with cell health, and researchers have developed an automated method called AutoMitoC to estimate this concentration quickly and easily. Genetic analysis identified 71 loci related to changes in mitochondrial DNA concentration, shedding light on potential links to health conditions like dementia.

Mitochondrial Dysfunction as a Hallmark of Environmental Injury

Carolina Duarte-Hospital et al.

Summary: Environmental factors have a significant impact on human health, including diet, sedentary lifestyle, and exposure to pollutants. The cellular and molecular events triggered by exposure to environmental pollutants vary greatly depending on age, chronicity, and exposure doses. Mitochondrial dysfunction is increasingly recognized as a major characteristic of environmental damage. This article reviews the multiple mechanisms by which pollutants cause mitochondrial toxicity in relation to chronic diseases and proposes the Aryl hydrocarbon Receptor (AhR) as an exposome receptor model that activates various toxic events through mitochondrial dysfunction. Finally, some remarks are provided on mitotoxicity and risk assessment.

Epigenome-wide DNA methylation signature of plasma zinc and their mediation roles in the association of zinc with lung cancer risk

Hua Meng et al.

Summary: This study investigates the role of DNA methylation in the association between zinc and lung cancer. The researchers identify several CpG sites associated with zinc levels in the blood and their potential impact on lung cancer risk. The results suggest that DNA methylation plays a biological role in the inverse association between zinc and lung cancer. Adding methylation risk scores improves the diagnostic capability for lung cancer.

Summary: Rubidium may have more pronounced effects on lowering blood pressure levels than potassium. High urinary rubidium concentrations are significantly associated with reduced risk of hypertension and lower blood pressure levels. Moreover, urinary rubidium concentrations are negatively associated with risk factors for hypertension.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Mitochondrial copper depletion suppresses triple-negative breast cancer in mice

Liyang Cui et al.

Summary: The research developed a mitochondria-targeted copper-depleting nanoparticle that effectively inhibited the growth of triple-negative breast cancer cells and improved survival rates in mouse models. By depriving copper within cancer cells, this approach induced metabolic changes and apoptosis, highlighting its potential clinical relevance.

Deciphering the genetic and epidemiological landscape of mitochondrial DNA abundance

Sara Hagg et al.

Summary: This study investigated the factors influencing mitochondrial DNA (mtDNA) abundance, revealing that it is higher in women compared to men and negatively correlated with age, smoking exposure, BMI, frailty index, and certain blood biochemistry markers. Genome-wide association study identified 50 regions significantly associated with mtDNA abundance, indicating the involvement of genes related to immunity, cancer, and mitochondrial function. The SNP-heritability of mtDNA abundance was estimated to be around 8%, and mtDNA abundance was found to be associated with leukemia, hematologic diseases, and hypertension.

Anna R. Smith et al.

Summary: This study evaluated the individual and joint associations of prenatal metals with mitochondrial DNA copy number and telomere length in maternal and cord blood. The results showed that maternal magnesium was associated with lower mtDNAcn, lead was associated with higher maternal mtDNAcn, and selenium was linked to longer cord blood TL. Nonlinear relationships and interactions were also observed among certain metals and biomarkers.
