The enzyme-modified comet assay: Past, present and future

Damian Muruzabal et al.

Summary: The enzyme-modified comet assay has been developed to detect various types of DNA lesions beyond the standard version, and has applications in human biomonitoring, environmental toxicology, genotoxicity testing, and basic research. Twelve enzymes have been employed, with some being extensively used while others less so. Additional enzymes could potentially expand the range of detectable DNA lesions and play a crucial role in understanding the mechanism of genotoxins.

In vitro and integrated in vivo strategies to reduce animal use in genotoxicity testing

Katherine Groff et al.

Summary: The unprecedented interest in implementing human-relevant, mechanistic in vitro and in silico testing approaches has been driven by scientific, financial, and ethical factors. The strategy outlined for assessing genotoxicity involves standard in vitro methods followed by advanced 3D tissue models, with the aim of reducing animal use, study time, and costs. Regulatory agencies encourage the adoption of these new strategies to advance toxicology testing practices.

Comet assay: a versatile but complex tool in genotoxicity testing

Eugenia Cordelli et al.

Summary: The comet assay is a versatile method for measuring DNA strand breaks in individual cells, with modified versions able to detect oxidized DNA bases and reveal DNA damage induced indirectly by reactive oxygen species. Despite being semi-quantitative, there are possible approaches to improve DNA damage quantitation and enhance protocol standardization for comparability. The in vivo comet assay has an advantage over the micronucleus erythrocyte test as it can be applied to any organ and has been used in risk assessment for specific food contaminants.
