Precise druggability of the PTH type 1 receptor

Ieva Sutkeviciute et al.

Summary: By using molecular dynamics simulations and elastic network model-based methods, the study identified a key mechanical site that modulates the collective dynamics of PTHR and successfully identified selective small molecules with strong negative allosteric properties for PTHR signaling. This computational pipeline provides a way to detect precise druggable sites and identify allosteric modulators for GPCRs, expediting the discovery of small molecules as novel therapeutic candidates.

Effector membrane translocation biosensors reveal G protein and βarrestin coupling profiles of 100 therapeutically relevant GPCRs

Charlotte Avet et al.

Summary: This study presents a set of BRET sensors to monitor the activation of G protein subtypes without modifying the receptors or G proteins. Profiling 100 therapeutically relevant human GPCRs resulted in pathway-specific concentration-response curves, revealing a diversity of coupling profiles.

Biased GPCR signaling by the native parathyroid hormone-related protein 1 to 141 relative to its N-terminal fragment 1 to 36

Karina A. Pena et al.

Summary: This study investigates the signaling pathway and biased signaling properties of native PTHrP protein hormone acting with PTHR. Using fluorescence imaging approaches, the study demonstrates that PTHrP(1-141) displays biased agonist signaling properties that are not mimicked by PTHrP1-36. The molecular basis for this biased signaling difference is attributed to the stabilization of a singular PTHR conformation and PTHrP(1-141) sensitivity to heparin.

Evaluations of daily teriparatide using finite-element analysis over 12 months in rheumatoid arthritis patients

Kumiko Ono et al.

Summary: In this study, daily teriparatide treatment was found to significantly increase bone mineral density (BMD) and predicted bone strength (PBS) in rheumatoid arthritis patients with osteoporosis. Increases in N-terminal type I procollagen propeptide (PINP) levels at 1 and 3 months were significantly associated with increased PBS at 12 months, with a threshold value of 75 μg/L at 1 month for predicting PBS increases at 12 months.

Compartmentalized GPCR Signaling from Intracellular Membranes

Stephanie E. Crilly et al.

Summary: GPCRs are integral membrane proteins that transduce signals from both the cell surface and intracellular membrane compartments, producing functional effects that differ from canonical plasma membrane signaling. The activity and signaling of GPCRs are influenced by intracellular membranes and their associated lipids and proteins, resulting in a spatial bias in GPCR signaling.

Summary: The study presents the structure of PTH2R bound to its endogenous ligand TIP39 and a heterotrimeric Gs protein, revealing the unique conformation of TIP39 and its mechanism of action on the receptor. It also identifies the basis of ligand specificity relative to TIP39, PTH, and PTH-related peptide, as well as the essential role of the N terminus in PTH2R activation. Additionally, a disease-associated mutation G258D significantly reduced cAMP accumulation induced by TIP39.

Will18F-fluorocholine PET/CT replace other methods of preoperative parathyroid imaging?

Luca Giovanella et al.

Summary: Cervical ultrasound and parathyroid scintigraphy are commonly used in diagnosing PHPT, but four-dimensional CT and radiolabeled choline PET have emerged as useful alternatives when initial imaging methods are inconclusive. Radiolabeled choline PET/CT shows excellent diagnostic performance and low radiation exposure, making it an ideal candidate to replace current imaging procedures for preoperative localization of HP in patients with PHPT, including challenging cases of postoperative persistent/recurrent disease.
