Oxidative Damage Blocks Thymineless Death and Trimethoprim Poisoning in Escherichia coli

Summary: Thymineless death (TLD) is a chromosome-based phenomenon that occurs in cells unable to synthesize dTTP. Recent studies proposed oxidative damage as a key contributor to TLD, but our experiments showed that blocking Fenton's reaction had no impact on TLD, while increasing oxidative damage completely prevented TLD. Therefore, oxidative damage does not play a role in TLD.

This oxygen-sensitive pausing during replication may serve as an adaptation to prevent replication under potentially lethal or mutagenic conditions.
