Procyanidins inhibit zearalenone-induced apoptosis and oxidative stress of porcine testis cells through activation of Nrf2 signaling pathway

Summary: This study assessed the effects of procyanidins (PC) on cell apoptosis caused by zearalenone (ZEA) and validated the role of Nrf2 in this process. The findings demonstrated that PC could enhance the cell antioxidant capacity, mitigate ZEA-induced cell apoptosis, and promote the expression of Nrf2 and its downstream genes. This discovery contributes to the clinical application of PC in preventing reproductive toxicity caused by ZEA.

Summary: This study investigates the role of the V0d1 subunit of V-ATPases in gut homeostasis. Knockdown of VhaAC39-1 in Drosophila leads to the loss of midgut acidity, increased gut microbiota, and impairment of intestinal epithelial integrity. It also causes the hyperproliferation of intestinal stem cells and activation of immune signaling pathways, resulting in overall disturbance of gut function and decreased lifespan in flies.

Summary: The study demonstrated that Sestrin protects Drosophila midgut from HgCl2-induced oxidative damage by inhibiting ROS production and stimulating the tissue regeneration program under regulation of the JNK signaling pathway. Knockdown of Sestrin in Drosophila midgut aggravated the disruption of intestinal organization induced by HgCl2, worsening ROS production and cell apoptosis. Additionally, JNK signaling was found to mediate Sestrin expression in the midgut.

Recent research has cast doubt on this hypothesis, including the effects of vitamin E on enzyme activities, signaling cascades, gene expression, and biomembrane structure. Overall, our knowledge of vitamin E's mechanism of action remains incomplete, with discussions on its metabolism, bioactivity of its metabolites, and implications for future clinical trials.

Zearalenone Exposure Disrupts Blood-Testis Barrier Integrity through Excessive Ca2+-Mediated Autophagy

Summary: The study demonstrated that exposure to Zearalenone (ZEA) resulted in severe testicular damage and a decrease in the expression of blood-testis barrier (BTB) junction-related proteins in a time-dependent manner, accompanied by an increase in autophagy-related proteins. In vitro, ZEA treatment led to an increase in cytoplasmic Ca2+ levels, autophagy markers LC3-II and p62 levels, and a decrease in BTB junction proteins, with the dislocation of the gap junction protein Cx43. Inhibition of autophagy or cytoplasmic Ca2+ was able to mitigate the effects of ZEA on BTB in TM4 cells.

The exact underlying mechanism of Zearalenone-induced toxicity in the reproductive system remains largely unknown, but theories of estrogen-like effects and oxidative stress damage show promise.
