Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Summary: The incidence of early-onset colorectal cancer (CRC) is increasing, and decisions on optimal adjuvant therapy should consider treatment adherence, adverse events, and expected outcomes. Compared to later-onset CRC, early-onset CRC patients had better performance status, higher adherence to treatment, and higher administered treatment intensity. However, despite these advantages, young age is negatively prognostic in high-risk stage III CRC, with significantly higher relapse rates and worse cancer-specific outcomes.

Blocking PLK1 genetically and pharmacologically increases sensitivity to oxaliplatin. CDC7 is identified as a critical downstream effector of PLK1 signaling and its inhibitor enhances the anti-tumor effect of oxaliplatin in CRC models.

WD40 repeat 43 mediates cell survival, proliferation, migration and invasion via vimentin in colorectal cancer

Summary: This study investigated the role of WDR43 in colorectal cancer, showing that its high expression is associated with increased tumorigenesis. Knockdown of WDR43 led to decreased proliferation, migration, invasion, and increased apoptosis in CRC cells. Additionally, the study found that WDR43 knockdown inhibited VIM expression and overexpression of VIM partially reversed the effects of WDR43.
