Nuclear Receptor-Mediated Hepatomegaly and Liver Regeneration: An Update

Pengfei Zhao et al.

Summary: Nuclear receptors (NRs) play critical roles in hepatomegaly and liver regeneration, indicating the potential of NRs as targets to promote liver repair after liver injury. This paper reviews the characteristics and molecular mechanisms of NRs in regulating hepatomegaly and liver regeneration, providing more evidence for NRs in the treatment of related liver diseases.

Adenosine-to-Inosine RNA Editing and N6-Methyladenosine Modification Modulating Expression of Drug Metabolizing Enzymes

Masataka Nakano et al.

Summary: Interindividual differences in drug metabolizing enzymes can affect the efficacy and adverse effects of drugs. Transcriptional regulation, epigenetic regulation, and post-transcriptional regulation are known mechanisms of variability. Recent discoveries have identified adenosine-to-inosine RNA editing and N-6-methyladenosine as novel regulators of drug metabolism. Understanding these mechanisms is crucial for understanding individual variability in drug response.

Molecular Regulation of Bile Acid Homeostasis

Supratim Choudhuri et al.

Summary: Bile acids play a crucial role in the digestion and absorption of dietary fats and fat-soluble vitamins, and they also impact fat, glucose, and energy metabolism. This review highlights the regulation of bile acid synthesis, transport, and homeostasis, as well as the importance of bile acids in modulating intermediary metabolism. Understanding these mechanisms has potential implications for the development of treatments for hepatic and intestinal diseases, as well as metabolic syndrome.

Mechanisms by Which Inducers of Drug Metabolizing Enzymes Alter Thyroid Hormones in Rats

Nichole R. Vansell

Summary: Increased disposition of thyroid hormones by xenobiotics can disrupt thyroid homeostasis and lead to thyroid follicular adenoma/carcinoma in rats. While induction of thyroxine (T4) glucuronidation by UDP-glycosyltransferase (UGT) enzymes is a common mechanism, other drug processing genes and proteins, such as transporters, are also involved in this process. Assessing the transport of thyroid hormones and their glucuronidation may provide a better indication of an inducer's potential in disrupting thyroid hormones.

The Complicated Role of Nuclear Factor Erythroid-Derived 2-Like 2 in Allergy and Asthma

Cheryl E. Rockwell et al.

Summary: Nuclear factor erythroid-derived 2-like 2 (Nrf2) is an important immu-nemediator that regulates various immune cell types in inflammatory diseases such as allergy and asthma. There is significant interest in targeting Nrf2 as a drug for inflammation, but the complexity of Nrf2 in the immune system complicates this. This review focuses on the role of Nrf2 in asthma and allergy, including regulating immune cell function and detoxifying xenobiotics that worsen these diseases.

Shiwei Zhou et al.

Summary: Facilitated transport is necessary for large size, charged, and/or hydrophilic drugs to cross the membrane. SLC drug transporters play crucial roles in drug transport and distribution in the human body, with high expression in major organs like the intestine, liver, and kidney. Understanding the transcriptional regulation of these critical drug transporters is essential for interpreting pharmacokinetics, pharmacodynamics, and improving drug therapeutic effectiveness.

Effect of Chronic Cadmium Exposure on Brain and Liver Transporters and Drug-Metabolizing Enzymes in Male and Female Mice Genetically Predisposed to Alzheimer's Disease

Hao Wang et al.

Summary: Cadmium exposure is associated with increased risks of Alzheimer's disease. ApoE4 gene is related to increased risks of AD. This study investigates the effects of Cd, ApoE, and sex on drug-processing genes and finds that Cd dysregulates these genes in a sex- and ApoE-genotype-specific manner, which may contribute to the variance in susceptibility to Cd neurotoxicity.

Summary: This study investigates the drug JNJ-2482272 as an anti-inflammatory agent and finds that it acts as an autoinducer in rats. In vitro experiments demonstrate that JNJ-2482272 activates the aryl hydrocarbon receptor (AhR) and induces the expression of cytochrome P450 and UDP-glucuronosyltransferase enzymes in liver cells, leading to the formation of metabolites.
