Can the interplay between autophagy and apoptosis be targeted as a novel therapy for Parkinson's disease?

Minke Bekker et al.

Summary: The development of effective treatments for Parkinson's disease relies on understanding the mechanisms of neurodegeneration. Autophagy and apoptosis are two cellular death pathways believed to play key roles in PD, with molecular overlap between them. Important proteins like Bcl-2 can suppress autophagy and apoptosis by inhibiting key molecular components such as Beclin-1 and Bax. Activation of apoptosis is mediated by molecules like JNK and p38, which increase the Bax/Bcl-2 ratio. Additionally, Akt can act as both an inducer of autophagy and an inhibitor of apoptosis by regulating Bax expression.

Activation of MAP3K DLK and LZK in Purkinje cells causes rapid and slow degeneration depending on signaling strength

Yunbo Li et al.

Summary: The conserved MAP3Ks DLK and LZK play critical roles in Purkinje cells, with DLK activation leading to rapid death and LZK activation causing slow degeneration. Each kinase induces JNK activation and apoptosis through distinct pathways. Deletion of CELF2 significantly attenuates LZK-induced Purkinje cell degeneration. Controlling the activity levels of DLK and LZK is crucial for neuronal survival and health.

Defined Tau Phosphospecies Differentially Inhibit Fast Axonal Transport Through Activation of Two Independent Signaling Pathways

Sarah L. Morris et al.

Summary: Research shows that Tau protein is phosphorylated at multiple sites, with some associated with tau pathology. The AT8 antibody can detect a complex phosphoepitope site that significantly increases in Alzheimer's disease and other tauopathies. Phosphorylation events may impact tau conformation, promoting activation of different signaling pathways, which in turn affect axonal transport.

Lost in Translation? Measuring Diabetic Neuropathy in Humans and Animals

Heung Yong Jin et al.

Summary: The global diabetes epidemic affects nearly 500 million people, damaging multiple organ systems with the blood vessels, eyes, kidneys, and nervous systems being particularly vulnerable. Peripheral neuropathy, impacting over half of individuals with diabetes, currently lacks effective treatment. Research suggests that traditional nerve conduction studies in large myelinated fibers may not be the sole method for clinical research anymore.

Vincristine- and bortezomib-induced neuropathies - from bedside to bench and back

Stefanie Geisler

Summary: Vincristine and bortezomib are effective chemotherapeutics for hematological cancers, but they can cause peripheral neuropathy as a main side effect, leading to potential permanent disability. Research using various cellular and animal models has shown that bortezomib induces transcriptional programs in the dorsal root ganglia, resulting in neuroinflammatory pathways and central sensitization, while vincristine has direct toxic effects on axons. These findings provide insights into the molecular mechanisms of vincristine- and bortezomib-induced neuropathies and potential avenues for developing treatments to prevent or reverse them, ultimately improving patients' quality of life during and after cancer therapy.

Vincristine-Induced Peripheral Neuropathy in Childhood Acute Lymphoblastic Leukemia: Genetic Variation as a Potential Risk Factor

Qing-Yan Yang et al.

Summary: Vincristine (VCR) is a commonly used chemotherapy medication for childhood acute lymphoblastic leukemia, but its neurotoxicity limits its clinical application. Genetic variants may affect the incidence and severity of VCR-induced peripheral neuropathy (VIPN). Individualized treatment strategies may help diagnose and manage VIPN earlier, improving patient quality of life.

Vincristine-Induced Peripheral Neuropathy (VIPN) in Pediatric Tumors: Mechanisms, Risk Factors, Strategies of Prevention and Treatment

Silvia Triarico et al.

Summary: Vincristine-induced peripheral neurotoxicity (VIPN) is a common side effect in pediatric cancer patients undergoing vincristine chemotherapy, leading to potential complications such as treatment delays or discontinuation. The pathogenesis of VIPN involves complex mechanisms including damage to nerve cell endothelium, mitochondria, and microtubules, with both patient-related and treatment-related factors contributing to its development. Despite research into various drugs for prophylaxis and treatment, consensus on the best approach for VIPN management in pediatric oncology patients remains elusive.

Molecular basis of V-ATPase inhibition by bafilomycin A1

Rong Wang et al.

Summary: The study reveals the structural basis for Bafilomycin A1 inhibition of V-ATPase, showing that six Bafilomycin A1 molecules bind to the c-ring and disrupt the interactions between the c-ring and subunit a, thus preventing proton translocation.
