Heterogeneity in phenotype, disease progression and drug response in type 2 diabetes

Anand Thakarakkattil Narayanan Nair et al.

Summary: An analysis of newly diagnosed diabetes patients reveals the impact of phenotypic variation on disease onset, clinical outcomes, and drug response. The study shows that patients with different phenotypes have varying risks of diabetic complications and respond differently to different drugs. These findings are important for personalized treatment of diabetes.

Single-cell chromatin accessibility identifies pancreatic islet cell type- and state-specific regulatory programs of diabetes risk

Joshua Chiou et al.

Summary: By analyzing human pancreatic islet cells using single-cell ATAC-seq, different cell clusters and transcription factors that underlie lineage- and state-specific regulation have been identified, which helps prioritize type 2 diabetes risk variants. This study demonstrates the power of single-cell epigenomics in interpreting complex disease genetics and sheds light on potential mechanisms underlying T2D.

The impact of non-additive genetic associations on age-related complex diseases

Marta Guindo-Martinez et al.

Summary: This study utilized an extensive GWAS strategy called GUIDANCE to improve genotype imputation by using multiple reference panels, including the X chromosome and non-additive models. By applying this methodology, 94 genome-wide associated loci were identified for 22 age-related diseases, with 26 previously unreported loci. The study also revealed that 27.7% of these loci would have been missed if standard imputation strategies with a single reference panel and only testing the additive model were used.
