Pathological mechanisms and therapeutic strategies for Alzheimer's disease

Yaojun Ju et al.

Summary: Alzheimer's disease is a complex neurodegenerative disease characterized by synaptic dysfunctions and neurotransmitter deficiencies. The pathological features of Alzheimer's disease include changes in β-amyloid and tau proteins. Aging and other factors are also risk factors for the disease. Therefore, developing treatment strategies that target different pathological factors is crucial.

Tau Post-translational Modifications: Dynamic Transformers of Tau Function, Degradation, and Aggregation

Carolina Alquezar et al.

Summary: Post-translational modifications play a crucial role in regulating tau protein function, including phosphorylation and acetylation. While these modifications may regulate protein function under normal conditions, they can be hijacked in disease states. Limited understanding exists on how these modifications impact tau protein, but studies indicate their critical role in localization, interactions, and other aspects.

Glutathione in the Brain

Koji Aoyama

Summary: Glutathione (GSH) depletion in neurons is common in neurodegenerative diseases and can lead to neurodegeneration. Excitatory amino acid carrier 1 (EAAC1) plays a central role in regulating neuronal GSH production, making it a potential target for therapeutic strategies in these diseases.

Copper (II) binding properties of an octapeptide fragment from the R3 region of tau protein: A combined potentiometric, spectroscopic and mass spectrometric study

Bettina Diana Balogh et al.

Summary: The copper(II) complexes of a peptide fragment from the R3 domain of tau protein have been studied using various methods to determine their coordination modes. The versatile coordination of copper(II) with the peptide is attributed to the presence of two adjacent histidyl residues, resulting in different complex formations at varying metal-to-ligand ratios. CD spectroscopy indicates the presence of coordination isomers with similar concentrations.

A current view on Tau protein phosphorylation in Alzheimer's disease

Susanne Wegmann et al.

Summary: The functions of Tau in the central nervous system are regulated by various posttranslational modifications, with phosphorylation being widely recognized as a pathological hallmark in diseases like AD. However, the similarities and heterogeneity of phosphorylation patterns between physiological and pathological Tau make it challenging to identify specific modifications as therapeutic targets and biomarkers.

Thermodynamics and structural characterization of the nickel(ii) and zinc(ii) complexes of various peptide fragments of tau protein

Bettina Diana Balogh et al.

Summary: Nickel(ii) and zinc(ii) complexes of various peptide fragments of tau protein were studied and it was found that the histidyl residues of the N-terminal and R3 regions can effectively bind these ions. Nickel(ii) ions promote deprotonation and coordination in alkaline solutions, forming square planar complexes, while zinc(ii) hydrolysis can be promoted by the adjacent histidines of the R3 domain.

Summary: Alzheimer's disease has a significant impact on individuals and society as a whole, with a large number of patients in the United States and a potential for continued growth in the future. The value of unpaid caregivers has increased despite a decline in their numbers, and healthcare costs are on the rise. Racial and ethnic disparities in healthcare persist, highlighting the need for greater diversity in dementia care workforce and clinical trials.
